Objective: The aim of this study was to establish a real-time quantitative PCR(qRT-PCR)method for the detection of microRNA(miRNA)in esophageal squamous cell carcinoma(ESCC).The expression of miRNA-206 in ESCC patients was detected and analyzed.And to provide a new evidence for the clinical diagnosis of ESCC patients.And to explore its role in tumorigenesis and progression,as well as its expression in sera of patients with ESCC and normal people,and to provide new evidence for the clinical diagnosis of ESCC.Methods: Collected 38 cases of ESCC patients with serum and clinical data,who was viaited at the North Sichuan Medical College Hospital Cardiothoracic Surgery in March 2015~October 2015.All ESCC patients had not received any anticancer therapy.At the same time in the physical examination center to collect 31 cases of healthy patients serum,blood CEA indicators and basic clinical data.Fluorescent quantitative RT-PCR Was used to detect the expression levels of miRNA-206 in serum samples from 38 patients with ESCC.21 healthy people served as controls,ce-miR-394 as a reference gene.The differences in miRNA-206 expression levels were analyzed.Receiver operating characteristics(ROC)curves of the miRNA-206 were constructed by statistical software to assess diagnostic power for ESCC.According to the ROC curve analysis,cut off values in diagnosis of ESCC for miRNA-206 expression in serum were selected.The sensitivity,specificity of serum miRNA-206 in diagnosis of ESCC werecalculated,and compared with serum CEA.Results: 1.The relative expression level of serum mi RNA-206 in ESCC patients was significantly lower than that in normal healthy patients.The mean expression levels in serum of ESCC and normal healthy subjects were0.7288±0.1771 and 1.5146±0.2485,respectively.The difference was statistically significant(P=0.000).2.The areas under ROC curve of miRNA-206 and in diagnosis of ESCC were 0.754,suggesting that miRNA-206 expression of ESCC has a high diagnostic efficacy.The sensitivity and specificity of the diagnosis based on the ROC curve were67.7% and 73.7%,respectively.3.The sensitivity of serum CEA in the diagnosis of ESCC was lower than that of serum mi R-206,and the specificity was slightly higher.Conclusions: 1.The expression of miRNA-206 in esophageal squamous cell carcinoma was lower than that in normal people.2.The preliminary results of this study suggest that the expression of miRNA-206 in serum can be used as a molecular marker of ESCC for clinical diagnosis.
