Objective: To investigate the effects of Glucose-insulin-potassium on septic shock kidney damage.Methods: 30 New Zealand white rabbits were randomly divided into three groups.Lipopolysaccharide group(LPS group,n = 10)and polarization liquid treatment group(GIK group,n = 10)were intravenously injected with lipopolysaccharide(0.5 mg/kg)through ear marginal vein to create septic shock kidney damage models.Control group(N group,n = 10)were treated by injecting equivalent dose of saline through ear marginal vein.After the modeling was complicated,the GIK group was injecting with polarization liquid via ear marginal vein(5ml/kg/h),as the same speed as N group and LPS group were injected with saline solution.Vital signs for three groups were monitored;levels of Blood glucose,lactic acid,blood potassium and Cystatin C were detected by extracting blood preparation before modeling,after modeling and at 1h,3h,6h after treatment respectively;All animals were sacrificed at 6h after treatment,levels of renal TNF-ɑ,MDA,and Caspase-3 level were detected.HE staining was conducted to observe pathological variations.Renal tubule mitochondrial injury was observed by electron microscope.Results: 1.Vital signs : compared with those in N group(P<0.05),rabbits in LPS group had a series of changes including accelerated breathing,decreased mean arterial pressure,fervescence and so on.These changes in the rabbits of GIK group were less serious than those in LPS group(P<0.05).2.Blood gas analysis: compared with those in N group,blood glucose,potassium and lactic acid increased,while PCO2 decreased in LPS group(P<0.05);compared with LPS group,GIK group was detected with lower levels of lactic acid and potassium(P<0.05).3.Renal injury indicators:TNF-ɑ,Cystatin C and MDA increased greatly in LPS group compared with those in N group(P<0.05)and the indicators as above were increased in GIK group but were still higher than those in N group(P<0.05);HE staining indicated that renal injury were relieved in GIK group compared with that in LPS group but was still more serious than that in N group.4.Apoptosis indicators: Caspase-3 expression of GIK group was down-regulated compared with LPS group(P<0.05),and was up-regulated compared with N group(P<0.05).5.Electron microscope: Mitochondrial was swelled obviously in renal tubular epithelial cell in LPS group compared with N group,while mitochondrial structure was clearly visible in GIK group performed better at swelling degree and without obvious membrane fracture.Conclusion: GIK has protective effects in rabbits of septic shock renal injury models by intravenous injected with LPS,the protective effects were through relieving mitochondrial injury and reducing renal tissues apoptosis in rabbits.
