Objective To syudy angiopoietin-1(ANGPT1)mRNA and protein expression in lung adenocarcinoma and its correlation,and to analyze the relationship between the clinicopathologic features and prognosis of lung adenocarcinoma.Methods The samples from surgical specimens,including lung adenocarcinoma tissues?their adjacent normal tissues and normal tissues which was collected between 2015-5 and 2016-7,all the samples were divided into two parts,one part reverse them into reverse transcription of DNA(cDNA),then using fluorescence quantitative PCR to detect the expression of ANGPT1 at the RNA level,another part was detected the expression of ANGPT1 at the protein level by immunohistochemistry SP method;also to explore the expression and clinicopathological significance.At the same time,extract the expression of ANGPT1 was detected in lung adenocarcinoma tissues?their adjacent normal tissues between 2012-7 and 2015-1 by immunohistochemistry SP method,analyse the relationship of their expression with clinicopathologic features.Meanwhile,analyse their correlation and make prognosis analyse.Results Study on the expression in RNA level showed that ANGPT1 mRNA in lung adenocarcinoma was lower than that of adjacent normal tissues,the expression level of ANGPT1 mRNA in adjacent normal tissues was higher than that of lung adenocarcinoma,high about 10.19 times,the difference was statistically significant(P < 0.05);the expression of ANGPT1 mRNA in normal tissues was 2.66 times higher than that in adjacent normal tissues,the difference was statistically significant(P < 0.05);the expression of ANGPT1 mRNA was not correlated with gender?age and differentiation(P > 0.05),and lymph node metastasis and TNM staging was related;low expression of ANGPT1 mRNA often leads to more tendency of late stage and lymph node metastasis.The expression of ANGPT1 protein in lung adenocarcinoma tissues,adjacent normal tissues and were 73/96(73)% and 21/107(19.6%),the negative expression of ANGPT1 in lung adenocarcinoma was significantly lower than that in paracancerous tissues and normal tissues,and the difference was statistically significant(X2=64.77,P=0.000),and paracancerous tissues and normal lung tissues without obvious differential expression;No differences in paracancerous tissues and normal lung tissues;the expression of ANGPT1 was related to lymph nodes ? TNM stage ? differentiation levels of the organization and vascular tumor emboli in the study of clinicopathological significance(P<0.05);For the Kaplan-Meier study,ANGPT1 positive group was lower than that of negative group(P<0.01);Cox regression analysis showed that lymph node metastasis,the positive expression of ANGPT1 TNM stage and vascular tumor emboli can be used as an independent prognostic factor in patients with lung adenocarcinoma.Conclusion ANGPT1 expressed the adverse trend in RNA and protein level,and high expression of ANGPT1 protein may be related to tumorigenesis?progression?invasion and metastasis of lung adenocarcinoma may serve as apotential therapeutic target in the treatment of lung adenocarcinoma.At the same time,there was a strong correlation between two indexes and prognosis,and it was may found to be an independent prognostic factor.
