IL-21 And Bcl-6 Regulates The Proliferation And Secretion Of Tfh And Tfr In The Intestinal Germinal Center Of Inflammatory Bowel Disease

Object: To investigate the express of IL-21 and Bcl-6,which effect on intestinal inflammatory bowel disease germinal center follicular helper T cells and follicular regulatory T cells,and the relationship between its expression and clinical features of inflammatory bowel disease.Methods:The blood samples of 134 patients and the intestinal tissues of121 patients with inflammatory bowel disease(IBD)were collected from the First Affiliated Hospital of Guangxi Medical University and diagnosed by pathology.In addition,80 cases of blood samples and 83 cases of intestinal tissue samples were collected from normal control group.The expression was detected by RT-PCR in patients with inflammatory bowel disease and healthy peripheral blood cytokines IL-21 and Bcl-6 mRNA;Using flow cytometry in patients with inflammatory bowel disease and healthy population in the peripheral blood of T follicular helper cells and follicular regulatory T cell proportion;Usingtriple-mmunofluorescence staining analysis of the distribution characteristics of intestinal tissue of patients with inflammatory bowel disease have follicular helper T cells in germinal center and follicular regulatory T cells.Results:1.The expression in patients with inflammatory bowel disease and healthy peripheral blood cytokines of IL-21?Bcl-6mRNA in Ulcerative colitis and Crohn disease were up-regulated distinctly in the control group(p<0.05 for all).2.contrast with this result,the flow cytometry show that compare with the control the level of Th17 cells?Tfh in the peripheral blood of UC group and CD group:There was increase in the percentage of Treg cells and follicular t cell.the differece was statistically significant(p<0.05 for all);The level of Treg cells?Tfr in the peripheral blood of UC and CD patients:There was increase in the percentage of Treg cells and Tfr.the differece was statistically significant(p<0.05 for all).3.Triple-mmunofluorescence staining show that the number of Tfh in the intestinal germinal center was obviously up-ragulated in the UC group and CD group compare with the control group(p<0.05 for all),whereas the Tfr in the intestinal germinal center was reduced in the UC group and CD group compare with the control group(p<0.05 for all).Conclusion:Bcl-6 and IL-21 can regulate Tfh/Tfr in the intestinal germinal center and promote the occurrence and development of inflammatory bowel disease...
