Objective: To explore a new idea for the comprehensive utilization of Chinese Herbal Medicine Plant Platycodon grondiflonm(Jacq.)A.DC.,to search for the characteristics compounds which could stand for composition in different harvest periods of Platycodonis Radix,and to solve the problem of how to recycle the offal come from the process of planting and harvesting perennial medicinal plant Platycodon grandiflorum.Methods: The HPLC fingerprints of 25 batches of different harvest periods Platycodonis Radix were established,and the characteristic compounds of Platycodonis Radix were collected by principal component analysis;The extraction process of total saponins from the non-medicinal parts of Platycodon grandiflorum was optimized by central composite design and response surface methodology;The single factor investigation was used for total saponin purification process;through prescription screening and process optimization,the total saponin was manufacture into nebulizer which used for atomizer and the method for the determination of characteristics compound Platycodin D was been built.Results: Including Platycodin D and four unknown compounds were found by principal component analysis from 55 characteristic peaks,and those five compounds could regard as characteristics compounds in different harvest periods of Platycodonis Radix;Optimum extraction parameters of non-medicinal parts of Platycodon grandiflorum were as follows: water absorption 1.82 m L·g-1,liquid material ratio 20.59 m L·g-1,soaking time 1.06 h,extraction times 3 times,extraction time 1.56h;Purification process parameters of total saponin were as follows: adsorption velocity 6BV·h-1,eluent 70% ethanol,elution velocity 3BV·h-1,eluent consumption 9BV;in these cases the total saponin purity was 62.91%,and the content of Platycodin D was 5.29%;1000 milliliters nebulizer which used for atomizer contain PGS 5g,oleum aurantii 0.2m L,1,2-propanediol 50 m L and each milliliter nebulizer content Platycodin D was more than 0.15 mg.Conclusion: The method combining HPLC fingerprint of Chinese medicine and principal component analysis has certain reference value in searching for the characteristic components of Chinese Medicine;the Platycodonis Radix total saponin process parameters could be optimized by central composite design and response surface methodology,and the obtained process can be used for the production of high purity extractive from Platycodon grandiflorum non-medicinal parts;the research process of Platycodonis Radix preparation nebulizer worked out new countermeasures for Chinese medicine comprehensive utilization.
