Objective: This study was based on the clinical treatment GSQGF deficiency and Blood Stasis primary osteoporosis efficacy and animal experiments GSQGF underlying bone mineral density in ovariectomized rats,combined with stem cell research experiments to explore GSQGF of bone marrow mesenchymal stem cell proliferation and osteogenic differentiation,the initial mechanism of action of anti-osteoporosis clear that party.Provide new experimental evidence for osteoporosis treatment of Traditional Chinese Medicine Compound.Methods:(1)In this study,bone marrow adherent culture isolated purified,establish inter-cultured SD rat bone marrow mesenchymal stem cell system,multi-directional differentiation analysis to identify the cell morphology and cell by cell.GSQGF(2)Preparation of different doses of saline containing serum and serum by MTT assay GSQGF containing serum on bone marrow mesenchymal stem cells.After osteogenic induction,alkaline phosphatase activity assay and alizarin red staining to detect the impact GSQGF containing serum for mesenchymal stem cell osteogenic differentiation.(3)Real-Time PCR,detection GSQGF containing serum after bone marrow mesenchymal stem cells intervention,Runx-2,OPG gene expression.Mechanism of action of drugs initially clear.Results:(1)rat bone marrow mesenchymal stem cells microscopic morphology: whole bone marrow adherent culture method between the separation and purification of cultured bone marrow mesenchymal stem cells,the cells grew well in the main spindle cells,radial,spiral-shaped,more uniform shape.Test results(2)MTT assay showed: GSQGF containing serum groups compared with the saline group,GSQGF containing serum could promote BMSCs proliferation(P <0.05),GSQGF(H)containing serum on proliferation of SD rat BMSCs role in promoting the best.(3)after osteogenic alkaline phosphatase activity assay and alizarin red staining showed: GSQGF(H),GSQGF(M)on BMSCs osteogenic role in promoting better than saline group(P <0.05).High-dose group had better role in promoting SD rat BMSCs osteogenic differentiation in.By Real-Time PCR,detection GSQGF containing serum on bone marrow mesenchymal stem cells after the intervention,GSQGF(H),GSQGF(M)group Runx-2,OPG gene expression was significantly higher than saline group(P <0.05).Conclusion:(1)GSQGF containing serum can promote the proliferation of SD rat BMSCs;(2)GSQGF containing serum can promote osteogenic differentiation of rat BMSCs SD action;(3)GSQGF containing serum can promote Runx-2 and OPG gene expression.It may to some extent reflect GSQGF treatment of osteoporosis Mechanism and BMP signaling pathways and OPG / RANK / RANKL pathway related.
