Objective: By imposing acupuncture on heroin addiction rats Baihui and Dazhui,to understand the apoptotic status of the hippocampus and the ventral tegmental area of the heroin re-addicted rats,to observe the effect of acupuncture on the endoplasmic reticulum membrane affected the expression of PERK、IRE1m RAN and protein and the effect of PERK-activated CHOP on the pro-apoptotic response pathway and the IRE1-activated JNK pro-apoptotic response pathway,through acupuncture the heroin addiction rats Baihui and Dazhui acupoints.Analyzing the effect of acupuncture on the stress response of endoplasmic reticulum,to explore the neuroprotective effect of acupuncture on brain injury in heroin addicted rats.Methods: 64 SD rats were randomly selected from 4 groups: control group,model group,acupuncture group,drug group,16 rats in each group.The model of heroin addicted rats was established by 8 days of muscle injection,and the stage of heroin addiction was established.After administration of heroin,the time was 5 days,the natural withdrawal;the experimental stage is called detoxification.According to the exposure(addiction)→detoxified experimental way,cycle 3 experimental phases,the establishment of rat model of heroin relapse.The hippocampus and VTA tissue of rats were collected at thirty-ninth days.Brain tissues were collected from the hippocampus and VTA on experimental day 39.TUNEL method was used to examine the changes of cell apoptosis in the hippocampus and VTA region of rats.PERK,el F2 a,CHOP,IRE1,JNK gene expression using fluorescence quantitative RT-q PCR detection,PERK,el F2 a,CHOP,IRE1,JNK protein expression using immunohistochemical method detection.Results: 1、Under the microscope,it was found that the hippocampus and VTA of the control group were almost difficult to find the cell apoptosis,most of the nuclei were oval or round,and most of the staining was light blue.Compared with the control group,the number of apoptotic neurons in the hippocampus and VTA parts of the model group was significantly increased(P<0.01),and the apoptotic bodies formed by the decomposition of the nerve cells were also observed.Compared with the model group and the drug group,the apoptotic bodies in the hippocampus and VTA in the acupuncture group were decreased.After heroin relapse model,model group,acupuncture group and medicine group in the hippocampus and VTA parts of total number of positive cells increased significantly compared with the control group(P<0.01);compared with the model group,acupuncture group and medicine group the total number of positive cells decreased significantly(P<0.01);and the drug group,the total number of acupuncture group of positive nerve cells decreased significantly(P<0.05).2、The expression of hippocampus in model group and VTA,el F2 a,CHOP,PERK parts IRE1,JNKm RNA compared with the control group increased(P<0.01);compared with the model group,the expression of acupuncture group and medicine group in the hippocampus and VTA,el F2 a,CHOP,PERK parts IRE1,JNKm RNA decreased significantly(P<0.01,P<0.05);and the drug group,expression in hippocampus of rats in the acupuncture group PERK,el F2 a,IRE1m RNA decreased significantly(P<0.05),the expression of VTA in brain area of rats in the acupuncture group el F2 a,CHOP,IRE1,JNKm RNA decreased significantly(P<0.05),the difference was statistically significant.3 、 Compared with the control group,model group and VTA expression in the hippocampus,el F2 a,CHOP,PERK parts of IRE1 and JNK protein were significantly increased(P<0.01);compared with the model group,the expression of acupuncture group and medicine group in the hippocampus and VTA,el F2 a,CHOP,PERK parts of IRE1 and JNK protein decreased significantly(P<0.01,P<0.05);and drug group,expression of brain hippocampus of acupuncture group rats,el F2 a,PERK and VTA site of IRE1 protein was significantly decreased(P<0.05),the difference was statistically significant.Conclusion: 1、Long term heroin use can cause apoptosis in the hippocampus and VTA sites in rats,by giving the rats with acupuncture Baihui and Dazhui,can reduce the apoptosis of nerve cells,and has a protective effect on brain nerve cell injury.2 、 Acupuncture treatment can inhibit the up-regulation of endoplasmic reticulum transmembrane protein PERK,IRE1 m RNA and protein expression,as well as the regulation of activated CHOP,JNK pro-apoptotic response pathway,this may be through the acupuncture of rats Baihui and Dazhui points,regulating endoplasmic reticulum stress response,one of the important role of intervention of heroin brain injury.
