| Objective: The SD rats were to investigate the influence of the exercise’s effor t on 2,3,7,8-TCDD poisoned SD rats’ expression of Nrf2 and other related prote in.Methods: Forty male SD rats(7 weeks old)were adaptability feeding for one week,then divided into four groups randomly,respectively non-exercise control group(NC),exercise control group(EC),non-exercise toxic group(NT),exercise toxic group(ET).NT,ET groups received first 2,3,7,8-TCDD intra peritoneal injection dose respectively of 6.4μg/kg·bw.Then received 21% of the initial doses exposure for the next 7 weeks.NC,EC groups were received sa me quantity of corn oil.EC,ET groups swam with weight burdened on tails(5% body weight),five days a week,30 minutes per day.Killed all the SD rat s after 8 weeks and tested the body weight,wet liver weight,relative liver we ight,the activities of the AST,the activities of the ALT,the content of the Kea p1,the content of Nrf2,the binding activity of the Nrf2-ARE.Results: TCDD exposure have main effect on wet liver weight of SD rats(P=0.049);TCDD exposure have main effect on relative liver weight SD rats(P=0.049);Exercise have main effect on the activities of the AST(P=0.020),TCDD expo sure have main effect on the activities of the AST(P=0.001);Exercise have mai n effect on the activities of the ALT(P=0.001),TCDD exposure have main effe ct on the activities of the ALT(P=0.049);In the NC group and the EC group,t he hepatocytes of the rats in the NE group were arranged neatly and the cell structure was intact.The NT cells were disordered and infiltrated with inflamma tory cells.The cells in the NE group were slightly disordered and infiltrated with inflammatory cells;Exercise have main effect on the expression of the Nrf2(P=0.004);Exercise have main effect on the expression of the Nrf2 in nuclear(P=0.000),TCDD exposure have main effect on the expression of the Nrf2 in nuclear(P=0.015),Exercise and TCDD exposure have interaction effect on the e xpression of the Nrf2 in nuclear(P=0.024);Exercise have main effect on the bin ding activity of Nrf2-ARE(P<0.001),TCDD exposure have main effect on the binding activity of Nrf2-ARE(P<0.001),Exercise and TCDD exposure have intera ction effect of on the binding activity of Nrf2-ARE(P=0.043).Conclusion: 1.Exercise can alleviate TCDD-induced decrease of total Nrf2 pro tein content in liver;2.TCDD exposure can reduce the content of Nrf2 nucleo protein in liver of rats;exercise can alleviate TCDD-induced liver Nrf2 nuclear protein content decreased.3.Exercise can effectively reduce the liver damage caused by TCDD by up-regulating the expression of Nrf2 and its related prot eins. |
