Objective:1.To develop a multifunctional impact system and evaluate its function for animal injury model and fundamental research.2.To explore a new pancreatic trauma model in rats and evaluate trauma condition features based on different injury area.3.To study proliferation regularity and analysis self-recovery function of pancreas stem cells in a large area of trauma.Methods:1.The impact-or was based on the theory of energy storing device,multifunctional impact device and measuring equipment.The effectiveness and stability of this impact-or were tested.2.Rats were divided into two groups randomly.One group used 3cm2 impact tip,the other group used 6cm2 impact tip,we used the developed multifunctional impact-or to strike the opened pancreas for different-area injury in 400 kpa pressure.At 24 h after trauma,the survival rate,intra-abdominal structural changes,serum potassium and calcium ion concentration,amylase and lipase activities,and pathologic alterations of the pancreas were examined.3.Rats were divided into two groups randomly,one group accepted6cm2/400 Kpa pressure and the other group is control group.At 1d,2d,3d,7d after trauma rats were killed,then 12 h before being killed,rats were injected intraperitoneally with 100 mg/kg Brd U.We take pancreas tissue,pancreatic cells apoptosis and regeneration were examined by TUNEL and IHC,PDX-1 was detected by RT-PCR,IHC and Western blot.Results:1.We successfully developed the multifunctional impact-or,of which maximum impact stress continuously adjusted from 0 to 200 kg.The compressive stress and extrusion stress of the system also could be continuously adjusted from 0 to 100 kg.The tests suggested that the effectiveness and stability of this device were favorable(P(27)0.05 and P(29)0.05).2.We successfully established the pancreatic trauma model in rats.Compared with the control group,the two injured groups(3cm2 and 6cm2)can formed obvious injury(p(27)0.05),and the 6cm2 injured group was worse(p(27)0.05).3.PDX-1 was rose major at 2d to 3d in pancreas trauma model and then drop to normal level.It's significantly different compared with the control group(p(27)0.05).Conclusions:1.This impact-or could be easily operated in variety ways,and its impact parameters were real-time detected.The effectiveness and stability of this device were successfully tested by tests and animal experiments.The impact-or could be used in different levels and modes of variety injury on animals.It is suitable for collision injury model and the related basic research.2.This pancreatic trauma model was easy and effective,and it can be operated repeatedly and stably.It is suitable for pathologic and therapeutic study of pancreas trauma.3.Pancreas stem cells were activated in the pancreatic trauma model,Then PSCs proliferated and differentiated other cells,which compensated the damage of pancreas.
