Objective: To compare the expression of GLUT1,PKM2 in BGC-823 cells after transfected with small interfering RN A of hypoxia-inducible factors(HIF-1??HIF-2?)and investigate the effect of HIF-1? and HIF-2? on the regulation of GLUT1?PKM2 in gastric cancer.Methods: Small RNA interference(RNAi)was used to silence BGC-823 cells.The mRNA and protein expression levels of HIF-1??HIF-2??GLUT1 and PKM2 were detected by quantitative real-time reverse transcription PC R and western blot.Results: Small RN A interference(RNAi)effectively silenced the expressions of HIF-1? and HIF-2? genes and downregulated the expression of GLUT1?PKM2(P < 0.05),and the levels of PKM2 decreased the most obvious when transfected with HIF-1? siRNA,(P > 0.05).There was no significant difference between the two groups in the regulation of GLUT1(P> 0.05).Conclusion: The results suggest that HIF-1? and HIF-2? are both involved in regulating GLUT1?PKM2.Compared to HIF-2? pathway,HIF-1? pathway play a major role in regulate PKM2 and thus they all contribute to the Warburg effect of gastric cancer.
