Objective:Compared with other tissue derived mesenchymal stem cells,synovial mesenchymal stem cells have a stronger ability to differentiate into chondrocytes.By co culture,using the interaction between cells to promote the differentiation of stem cells,combined with the composite scaffold to provide three-dimensional space is more conducive to cell differentiation.The purpose of this study is the synovial mesenchymal stem cells mixed with chondrocytes cultured in natural and artificial materials,observe the 3D environment of synovial mesenchymal stem cells repair mixed with cultured cartilage cells after the damage ability of articular disc.Methods: synovial tissue and cartilage tissue were obtained from SD rats,and the mesenchymal stem cells and chondrocytes were obtained by enzymatic digestion.Methods:SD rat synovial tissue and cartilage tissue were obtained by enzyme digestion method.The third generation of synovial mesenchymal stem cells and second generation chondrocytes,two to 1: 2 ratio of mixed culture loaded on chitosan collagen type /I natural scaffolds,to implanted subcutaneously in nude mice and cultured for 4 weeks,8 weeks,by immunohistochemistry.Results:Replantation of 4 weeks,8 weeks after stent materials to maintain the disc,damaged articular disc with chitosan scaffold close adhesion,the surface with a layer of fascia,HE staining showed that 8 weeks when cells enter inside the material at 4 weeks was significantly increased,the damage of articular disc and scaffold materials contact parts are connected by stromal cells,chemical detection immunohistochemistry,the positive expression of cells and cell matrix type II collagen proteoglycan positive expression of toluidine blue staining.Immunohistochemistry and toluidine blue staining results showed positive expression.Conclusion: The experimentalresults show that the composite scaffolds containing co cultured cells can repair damaged articular discs.
