Objective:To investigate the hydroxyapatite / tricalcium phosphate and allogeneic bone scaffolds compounding with rabbit adipose derived stem cells(rADSCs),and composite meterial effect repairing rabbit spine bone defect.Method:36 New Zealand white rabbits were randomly divided into 3 groups,each group includes 12 rabbits.The HA/?-TCA +rADSCs composite meterial were implanted in the bone defect,in the experimental group.In the control group,allogeneic bone+r ADSCs composite meterial were implanted in the bone defect.While,Blank control group without any treatment.After4,8,12 weeks of the surgery,X-ray was taken.And after 12 weeks of the surgery,all animals were put to death for gross observation of specimens and histopathological observation.Results: After 12 weeks of the surgery,the experimental group's X-ray and gross observation of specimens pointed out that the basic bone defect repair,no obvious boundary.The experimental group's histopathological results point out that there were more trabecular structures and osteoblasts at low magnification while there were more vesicular adipocytes in the bone marrow,Only a small amount of scaffolds,no obvious fibrous tissues at low magnification.The experimental group's X-ray and gross observation of specimens pointed out that there was a large amount of bone tissue formation and a small amount of fibrous tissue covered in the bone defect.And experimental group's histopathological results point out that the structure of allogeneic bone scaffold and the new bone tissue were observed at low magnification.While the blank control group's X-ray and gross observation of specimens pointed out that there was a small amount of callus formation in bone defect,mainly with a large number of fibrous tissue filling.And the blank control group's histopathological results point out that there was a large numberof fibrous tissues and a few trabecular bone structures were seen at low magnification.Conclusion:There has good biological compatibility of rADSCs in HA/?-TCA and allogeneic bone scaffolds,and both of composite meterials can be better repairing in spinal defects.Both of the composite meterials can be chosen as bone tissue engineering scaffold for repairing bone defect.
