Objective To investigate the influence of heart failure and the expression of HSP22 protein by inhibiting JAK/STAT signaling pathway with AG490.Methods Wistar rats were randomly divided into three groups:control group(n=15);heart failure group(n=15),narrowed abdominal aorta diameter up to 0.45mm;AG490 intervention group(n=15),administered with AG490 by intraperitoneal injection after abdominal aortic coarctation.Observed the survival condition after the operation,at 4 weeks and 8 weeks detected the interventricular septal depth(IVSd)? left ventricular posterior walldepth(LVPWd)? left ventricular end diastolic diameter(LVEDd)? left ventricular end systolic diameter(LVEDs)?left ventricular ejection fraction(LVEF)by echocardiography,and left ventricular mass index(LVMI)?heart/body ratio?lung/body ratio?BNP? the change of HSP22 protein expression were investigated after 8 weeks.Results At 4 weeks,the IVSd,LVPWd,LVEDs,LVEDd and EF in the heart failure group were significantly different from those in the control group and the AG490 intervention group(P<0.05).There was no statistically difference in IVSd,LVEDd and EF between the AG490 intervention group and the control group(P>0.05).At 8 weeks,there were significant differences in IVSd,LVPWd,LVEDs,LVEDd and EF between the 3 groups,the change of heart failure group was more obvious(P<0.05).Compared with the control group,LVWI,?heart / body ratio,?lung / body ratio in the heart failure group and AG490 intervention group were increased(P<0.05),those index in heart failure group increased significantly(P<0.05).Under optical microscope,myocardial fiberturbulent,cardiomyocytes edema in heart failure group,the degree of myocardial morphological changes in the AG490 intervention group were not as obvious as that in the heart failure group.The BNP in heart failure group was significantly higher than that in the other two groups(P<0.01),the BNP in AG490 intervention group was higher than that in the control group,and lower than that in the heart failure group,the difference was statisticallysignificant(P<0.05).There was no positive expression of HSP22 protein in the control group,the HSP22 protein mean density in AG490 intervention group was significantly higher than that in the heart failure group(P<0.01).Conclusion JAK/STAT signaling pathway involved in process of chronic heart failure,intervention of JAK/STAT signaling pathway can reduce the severity of heart failure of rats with abdominal aortic constriction.HSP22 may play a protective role in cardiac function.
