Objective: Alveolar Echinococcosis(AE)is a parasitic disease characterized by Echinococcus multilocularis(Em)parasitic in the humans with chronic invasive liver injury.The aim of this study was to observe the changes of the number and type of lesions in the liver by the time of different infection time,and to evaluate the damage degree of the liver in the mice under different infection doses.To investigate the changes of CD4 + T cell subgroups and the expression of inflammatory cytokines in different doses,and to analyze the effects of different immune states on the parasitism of Echinococcus multilocularis.Methods:(1)Five different doses of mice(50psc,250 psc,500psc,1000 psc,2000psc)were used to achieve the purpose of infection of Echinococcus Mice liver specimens were collected at 7 time intervals(2 weeks,4 weeks,8 weeks,12 weeks,16 weeks,20 weeks,24 weeks).HE staining was performed to calculate the number of liver lesions in mice at different time and dose type;(2)?-SMA,RS,ki67 staining were performed at 3 different time points(2 weeks,12 weeks,24 weeks),and liver fibrosis levels and lymphocytes were evaluated at 3 different doses of Em(50psc,500 psc,2000psc)proliferation level;(3)select three doses(50psc,500 psc,2000psc),three infection period(2 weeks,12 weeks and 24 weeks)in mice,separation of liver lymphocytes,detection of liver areathe change of CD4 + Tcell subgroups and related inflammatory cytokines gene expression level.So as to analyze the effect of different immune states on Em.Results:(1)The number of lesions produced by high dose(2000psc,1000psc)was significantly higher than that of low dose group(P <0.05).The number of lesions did not decrease with the prolongation of infection time.And,the lesion area is large,the liver is seriously damaged.However,low doses(50 psc,250 psc)and medium doses(500 psc)of Em were injected into mice.As the infection time prolonged,the lesion could be cleared by the host and local liver injury could also be restored;(2)The positive rate of ?-SMA and RS staining in high dose group was significantly higher than that in low dose group(P < 0.05).The expression of COL1A1 and COL3A1 at 2weeks and 24 weeks after 2000 psc dose was significantly higher than that of normal control group;(3)250 doses did not change significantly at different infection time points Th1/Th2,and 500 psc dose Th1/Th2 was significantly increased at 24 weeks of infection.The dose of Th1/Th2 was significantly decreased at 24 hours after 2000psc;(4)The high dose group and low dose group showed Treg/Th17 balance shifted to Th17 in late infection(24 weeks).Conclusion:(1)low-dose Echinococcus multilocularis infectde in mice,the resulting E.m removed by the host,and the high-dose Echinococcus multilocularis infection can not be removed after infection;(2)high-dose Echinococcus infection mice,can cause more Severe hepatic fibrosis;(3)host immune type bias Th1 and Th17 may be beneficial to the E.m lesion clearance.
