Objective:To observe the effect of BMSCs(Bone Marrow Mesenchymal Stem Cells)on cerebral infarction volume,neurological score,the cell apoptosis in the cerebral ischemia area after ischemia/reperfusion in rats,and research the underlying mechanism of neuroprotection of BMSCs by inhibiting neurocyte apoptosis.Method: separated,cultivated,identified and marked the BMSCs.60 SD male rats were divided randomly into sham group,cerebral ischemia-reperfusion model group and the the BMSCs experimental group,every group included 20 SD male rats.The model group didn't perform the occlusion of middle cerebral artery,the other groups occluded the middle cerebral artery using the thread embolizing,after 90 min ischemia followed by 24 h reperfusion,the cerebral ischemia/reperfusion models were established.The experimental group rats were injected with 1 ml of BMSCs by cauda vein,the concentration of the BMSCs is 1×109/L,the other groups were injected with 1 ml of saline by cauda vein.The neurological scores were calculated respectively on the firet day,the first week and the second week.The cerebral infarction volume were calculated on the second week.The expression of Bax/Bcl-2 m RNA were evaluated by RT-PCR,the expression of Bax/Bcl-2 m RNA were evaluated by Western-Blot,the expression of caspase-3 and Survivin protein were evaluated by Western-Blot.Results: Compared with the model group,the neurological scores,brain infarct volume(P<0.05)decreased;improve the expression of Bcl-2 m RNA and protein(P<0.05);inhibit he expression of Bax m RNA and protein(P<0.05).improve the expression of Survivin protein(P<0.05);inhibit he expression of caspase-3(P<0.05).Conclusion:1.BMSCs can reduce brain infarction volume and have the effect of neuroprotection.2.BMSCs can reduce the neurological deficit and promote the function of the nerve.3.Improve the expression of Bcl-2 m RNA and protein.4.Inhibit he expression of Bax m RNA and protein.5.Improve the expression of Survivin protein and inhibit he expression of caspase-3.In conclusion,BMSCs can inhibite neurocyte apoptosis,improve the ischemia/reperfusion tolerance of brain,but the underlying mechanism need further in-deph study.
