Objective:To study the wake-promoting action,improving learning and memory ability of Schizandrin.Method:1.To study the effect of the Schizandrin on spontaneous activity of mice.Select 40 ICR mice and divided them 4 groups randomly with 10 in each group.Divided them into blank group,low dose medium dose and high dose group.The administration group were given different concentrations of Schizandrin suspension liquid and the concentrations are 50mg/kg,100mg/kg,200mg/kg respectively(the volume of the gastric perfusion was 0.4ml/20g).In the blank group,the experimental animals were administered with 1%sodium carboxymethyl cellulose solution once through the intragastric administration channels(the volume of the gastric perfusion was 0.4ml/20g).The above groups of experimental animals after the administration of 20 min,put them into the independent activity tester,adapt to the environment for 10 min,then measured the number of independent activities and the number of stand times of experimental animals in the four groups in 10min.2.The effects of Schizandrin on the sleep of freely moving ratsSelect 24 SD rat and divided them 3 groups randomly with 8 in each group.Divided them into low dose medium dose and high dose group.The administration group were given different concentrations of Schizandrin suspension liquid and the concentrations are 50mg/kg,100mg/kg,200mg/kg respectively(the volume of the gastric perfusion was 1ml/100g).In the blank group,the experimental animals were administered with 1%sodium carboxymethyl cellulose solution once through the intragastric administration channels(the volume of the gastric perfusion was 1ml/100g).To use cortical electroencephalography(ECoG)and analysis technique,the EEG of the rats was recorded for 24 hours and analyzed EEG in two hours,The sleep latency,two hours accumulated sleep time and wake time,the cumulative percentage of each sleep phase(Wake%,NREMS%,REMS%)is observed,Schizandrin research rats sleep duration and impact of the sleep phase changes.3.To study the effect of Schisandrin a on the performance of Morris water maze test in ratsSelect 24 SD rat and divided them 3 groups randomly with 8 in each group.Divided them into low dose medium dose and high dose group.The administration group were given different concentrations of Schizandrin suspension liquid and the concentrations are 50mg/kg,100mg/kg,200mg/kg respectively(the volume of the gastric perfusion was 1ml/100g).In the blank group,the experimental animals were administered with 1%sodium carboxymethyl cellulose solution once through the intragastric administration channels(the volume of the gastric perfusion was 1ml/100g).The above groups of experimental animals after the administration of 30 min,then navigation test,recorded the escaping latency.On the fifth day the medication and dosage consistent with the first four days,after the administration of 30 min,then spatial probe test,recorded the escaping latency and the number of times a platform is found within the specified time.4.To study the effect of Schisandrin a on neurotransmitter of brain in ratSelect 24 SD rat and divided them 3 groups randomly with 8 in each group.Divided them into low dose medium dose and high dose group.The administration group were given different concentrations of Schizandrin suspension liquid and the concentrations are 50mg/kg,100mg/kg,200mg/kg respectively(the volume of the gastric perfusion was 1ml/100g).In the blank group,the experimental animals were administered with 1%sodium carboxymethyl cellulose solution once through the intragastric administration channels(the volume of the gastric perfusion was 1ml/100g).The above groups of experimental animals after the administration of 30 min,the animals were decapitated,then separated brain tissue,weighed and quick-frozen,after processing,HPLC with electrochemical detection method was determinated monoamine transmitters in rat brain.Results:1.To study the effect of the Schizandrin on spontaneous activity of mice.The test results shoued that,compared with the control group,the active and standing times of the experimental animals in the middle and high-dose groups of Schisandrin were significantly decreased(p<0.01).2.The effects of Schizandrin on the sleep of freely moving ratsCompared with the previous administration,Schisandrin low,medium and high dose groups could significantly prolong the sleep latency(p<0.01).In the sleep-wake phase,Schisandrin showed a dose-effect relationship within a certain range,especially in high-dose groups.It not only could significantly prolong the Wake time and the percentage of Wake within the unit time(p<0.01),reduce NREMS time and the percentage of NREMS per unit time(p<0.01),also could decrease the REMS and the percentage of REMS per unit time.Its effect could be maintained for 6 hours.The three dose groups of Schisandrin had no significant effect on the number of Wake(p>0.05),however,the high dose groups could reduce the number of occurrences of REMS and NREMS per unit time(p<0.05).3.To study the effect of Schisandrin a on the performance of Morris water maze test in ratsIn the Morris water maze location navigation experiment,with the increase in the number of training and administered days,until the fourth day of training,compared with the control group,the escape latency of the experimental animals in the low,medium and high dose groups of Schisandrin was significantly shortened(p<0.01).In the space exploration experiment,compared with the control group,the escape latency of experimental animals in the middle and high-dose group of Schisandrin was shortened,and the number of times they find the platform increased(p<0.01).4.To study the effect of Schisandrin on neurotransmitter of brain in ratCompared with the control group,The test results of monoamine neurotransmitters in different brain regions showed that The effect of Schisandrin on NE,DA and 5-HT levels expressed as dose-dependent in different brain regions.Schisandrin high dose groups could significantly increase hippocampus brain areas of NE,DA and 5-HT NE content and the content of the hypothalamus(p<0.01).The contents of NE,DA and 5-HT in striatum were not significantly affected by the three dose groups of Schisandrin(p>0.05)Conclusion:Schisandrin may increase the levels of NE in the hippocampus and hypothalamus,DA and 5-HT in the hippocampus,promoting wakefulness effect and impromving the ability of learning and memory.
