Purpose:through the rat ischemic myocardial mitochondrial permeability transition pore opening of the regulating mechanism in the study,to explore the molecular mechanism of acupuncture preconditioning against myocardial ischemia,and at the same time in this paper,the mitochondrial permeability transition pore opening and acupuncture pretreatment;At the same time as the provisions for clinical prevention and treatment of ischemic heart disease and mitochondria and the relationship between the electric acupuncture preconditioning against myocardial ischemia provide scientific basis for experiment.Methods:96 rats by random number table corresponding divided into blank group,model group,acupuncture preconditioning group(acupuncture shenshu point),ischemic preconditioning group.To live in the rat heart left anterior descending coronary artery ligation of copying myocardial ischemia and reperfusion injury model.Blank group:the former building,fixed with acupuncture preconditioning group rats for 30 minutes but not acupuncture;1 times per day,continuous 15;Building in the coronary arteries in thread but not ligation,the whole experiment process is not on other factors.Model group:the former building with fixed set of blank;Myocardial ischemia and reperfusion model,the whole experiment process is not on other factors.Acupuncture preconditioning group;15th made before the film began to acupuncture;Once per day,every time 30 min,a total of 15;Myocardial ischemia and reperfusion model.Ischemic preconditioning group:building before the same as the blank group;When building on coronary artery ligation to block blood flow 5 min before,perfusion 5 min.So repeated 3 times in a row,a total of 30 min ischemic preconditioning.Myocardial ischemia after the success of the building 0 h,24 h,48 h,serum CK,LDH,serum myocardial ischemia area and myocardial cell mitochondrial membrane permeability transition pore opening degree.Results:1.The model group myocardial infarction area of more than 50%,a significant rise in serum CK and LDH values,dye laser confocal microscope mitochondria green fluorescent compared with the blank group was obviously reduced,the experimental results show that the activity of mitochondrial membrane permeability transition pore was significantly reduced(P<0.01).2.With the model control group,ischemic preconditioning group and acupuncture preconditioning group immediately myocardial infarction area,serum CK and LDH were significantly reduced,laser confocal microscope showed that mitochondrial fluorescence intensity is significantly reduced,but in ischemic preconditioning group and acupuncture preconditioning group immediately the indicators no significant difference(P>0.05).3.Acupuncture preconditioning 24h,48h group and ischemic preconditioning group and acupuncture preconditioning group immediately controlled area of myocardial infarction(mi),serum CK,LDH,further reducing;And laser confocal microscope fluorescence intensity observed in mitochondria have certain enhancement(P<0.05 or P<0.01).4.Through the experimental data shows that acupuncture preconditioning group and ischemic preconditioning group can be caused by the treatment of myocardial ischemia reperfusion injury;And perhaps acupuncture preconditioning 24 h group is the best treatment plan.Conclusion:1.Acupuncture pretreatment can be used for clinical prevention and treatment of myocardial ischemia reperfusion injury,and has a good curative effect in clinical treatment.2.Electric acupuncture pretreatment in the different period of myocardial protection,there may be differences may vary with time and treatment effect is enhanced for the different peak-weakening trend.3.Acupuncture preconditioning and ischemic preconditioning can reduce serum CK,LDH,reducing reperfusion injury caused by myocardial infarction area,so as to reduce myocardial apoptosis.4.Acupuncture preconditioning may be by inhibiting myocardial ischemia reperfusion mPTP and opening up,in the process of reducing the mitochondrial apoptosis,can reduce the myocardial ischemia caused by injury.
