Objective:It will be researched that the changes of the energy metabolic rate of sleep-wake cycle under different light conditions on rats and the effects of Semen Ziziphi Spinosae freeze-dried powder and its mechanisms.Methods:1 Study on the Change of Energy Metabolic Rate in Rats with Awakening-Sleep Cycle(24h)under LD ConditionSix SD male rats with cortical EEG and EMG electrodes were placed in the metabolic cage of the Animal Monitoring System(CLAMS),one per cage.1 per cage.Under the condition of 12h light and shade alternating light(L/D:12h/12h,LD)and free drinking water,By irrigation route,given rats distilled water,as early as 7:00,for 7 days.The 24-hour wake-sleep cycle was recorded by rat cortical electroencephalography and CLAMS system activity evaluation module,indirect thermo-thermal module and feeding test module.(Metabolic rate)and other indexes were used to study the changes of energy metabolism in the wake-sleep cycle(24h).2 Study on the Change of Energy Metabolic Rate in Rats with Awakening-Sleep Cycle(24h)Twelve SD male rats with cortical EEG and EMG electrodes were randomly divided into LD group and DD group.(DD/24h:DD)and free drinking water(DD)were used in the metabolic cage of the animal monitoring system(CLAMS),respectively,in the 12h light and dark alternating light cycle(L/D:12h/12h,LD)Conditions,The rats in the LD group and the DD group were treated with distilled water,7:00 pm for 7 days,Using the cortical electroencephalographic method and the CLAMS system activity evaluation module,the indirect heat measurement module growth rate(metabolic rate)and body weight.3 Effect of jujube root freeze-dried powder on the change of energy metabolism rate in awakening-sleep cycle(24h)in DD patients12 SD male rats with cortical EEG and EMG electrodes were randomly divided into DD group and administration group,6 rats in each group,Were placed in the metabolic cage of the animal monitoring system(CLAMS).Under the condition of under DD condition.4 Effect of jujube root freeze-dried powder on energy metabolism in rats with awakening-sleep cycle(24h)Eighteen SD rats were randomly divided into LD group,DD group and administration group,6 rats in each group.The rats in the experimental animal detection system(CLAMS)were treated with LD and DD(LD group and administration group),and the corresponding experimental animal distilled water and jujube root freeze-dried powder solution were given as early as 7 days for 7 days,Remove the decapitated remove the thalamus.The leptin,POMC and NPY,which were involved in the regulation of energy in the hypothalamus,were measured by enzyme-linked immunosorbent assay(ELISA)to study the mechanism of the freeze-dried powder of Suanzaoren on the energy change in the wake-sleep cycle.Results:1.The changes of energy metabolism rate in rats during waking-sleeping cycle(24h)under LD conditions.The results show that the sleeping time,the waking time,the number of voluntary activities,the food ration and the quantity of heat produced all in a dynamic process during the waking-sleeping cycle(24h)under LD conditions.The total waking time,the number of voluntary activities,the food ration and the quantity of heat produced at night(12h)was higher than that in the daytime(P<0.01),but the total sleeping time was lower than that in the daytime(P<0.01).2.The changes of energy metabolism rate in rats during waking-sleeping cycle(24h)under DD conditions.The results show that under DD conditions,compared with LD conditions the sleeping time,the waking time,the number of voluntary activities,the food ration and the quantity of heat produced occurred obvious dynamic changes.Compared with the LD group,the W in the DD group has significant increase in the daytime(P<0.01),and the W has significant decrease at night(P<0.01)and the total W has significantly increased(P<0.01)during all day(P<0.01).The NREM has significant decrease in the daytime(P<0.01),The NREM has significant increase at night(P<0.01),and total NREM has significant decrease during all day(P<0.01);The REM has significant decrease in the daytime(P<0.01),the REM has significant increase at night(P>0.05),the total REM has significant decrease during all day(P<0.01).Compared with LD group,the waking time under DD conditions has significant increase in the daytime(P<0.01),the waking time increased significantly at night(P>0.05)and the waking time extremely significantly in the daytime(P<0.01);the sleeping time was significantly reduced in the daytime(P<0.01),the sleeping time significantly reduced at night(P>0.05)and the sleeping time in the daytime was significantly reduced(P<0.01).Compared with LD group,the number of spontaneous activity in the daytime under DD conditions was significantly increased(P<0.01),there was no significant difference in the number of spontaneous activities at night(P>0.05),the number of spontaneous activities in the daytime was significantly increased(P<0.01).Compared with the LD group,there was no significant difference in the food ration in the daytime(P>0.05)in the DD group,and there was a significant increase in the food ration at night(P<0.05)and the total food ration was significantly increased during all day(P<0.05).Compared with the LD group,the quantity of heat produced of the DD group have no significant difference in the daytime(P>0.05),and the quantity of heat produced decreased significantly at night(P<0.05)and the quantity of heat produced decreased significantly(P<0.05)during all day.Compared with the LD group,the weight of the experimental animals in the DD group increased significantly(P<0.05).3.The effect of Suanzaoren Freeze-dried Powder on the changes of energy metabolism rate in rats during waking-sleeping cycle(24h)under DD conditions.The results show that the waking time and the sleeping time,the number of voluntary activities,the food ration and the quantity of heat produced occurred obvious dynamic changes under the influence of Suanzaoren in the DD group;Compared with the DD group,the W of the experimental animals in the daytime decreased significantly in the DD group(P<0.05),the W at night increased significantly(P<0.05)and the W was significantly increased during all day(P<0.01),the NREM increased significantly in the daytime(P<0.05),the NREM had no significant difference at night(P>0.05)and the NREM increased significantly during all day(P<0.05),the REM increased significantly in the daytime(P<0.05),the REM increased significantly at night(P<0.05)and the REM increased significantly during all day(P<0.05).Compared with the DD group,the waking time of the drug group was significantly reduced in the daytime(P<0.05),the waking time had no significant difference at night(P>0.05)and the total waking time was significantly reduced during all day(P<0.05);the sleeping time was significantly increased in the daytime(P<0.05),there was no significant differences in the sleeping time at night(P>0.05)and the total sleeping time increased significantly during all day(P<0.05).Compared with DD group,the number of spontaneous activities of the drug group was significantly reduced in the daytime(P<0.05),the number of spontaneous activities at night there was no significant difference(P>0.05)and the number of spontaneous activities decreased significantly during during all day(P<0.05).Compared with the DD group,there was no significant difference in the food ration of the drug group between the drug group in the daytime(P>0.05),and the food ration decreased significantly at night(P<0.05)and the total food ration decreased significantly during all day(P<0.05).Compared with the DD group,the quantity of heat produced of the drug group increased significantly in the daytime(P<0.05),and the quantity of heat produced increased significantly at night(P<0.05)and the quantity of heat produced increased significantly during all day(P<0.05).Compared with the DD group,the weight of the experimental animals in the drug group decreased significantly(P<0.05).4.The effect of Suanzaoren Freeze-dried Powder on the changes of energy metabolism rate in rats during waking-sleeping cycle(24h).Compared with LD group,the leptin content of the DD group in the hypothalamus decreased significantly(P<0.01),the content of POMC decreased significantly(P<0.01),the content of NPY increased significantly(P<0.05);under the DD condition,compared with DD group,the leptin content of the drug group in hypothalamus increased significantly(P<0.01),the content of POMC increased significantly(P<0.01),the content of NPY decreased significantly(P<0.01).Conclusion:1.Under the LD condition,in the awakening-sleep cycle,the rats have an increased energy metabolism rate in the night(awakening)and have a decreased energy metabolism in the daytime(sleep).2.Under the DD condition,in the awakening-sleep cycle,the rats' awakening time increased,the sleeping time decreased and lack of sleep.At the same time,the rats' self-activity reduced,the food intake increased and the calorie production reduced.These cause their energy metabolic rate disorder and weight gain.3.Wild jujube kernel freeze-dried powder can correct the energy metabolism rate disorder in rats with insufficient sleep in DD condition.4.Wild jujube kernel freeze-dried powder can correct the energy metabolism rate disorder in rats with insufficient sleep by increase the content of leptin,POMC and reduce the content of NPY in the hypothalamus.
