Osteochondral tissue engineering has become an effective approach to cure joint injury due to its low immune response and better reconstruction of the original morphology,structure and function.The core of tissue engineering is to establish a three-dimensional cell-scaffold construct.This scaffold provides not only a place for nutrient transfer and cells'growth and metabolism,but also a basis for new tissue and organ reconstruction with proper morphology and function.However,there are still some limitations in scaffold design such as no rational methodology available for analysis of scaffolds,and the existing scaffold system seems to lack the so-called vertical gradient function.This thesis has two main objectives:(1)to establish a design methodology to make the investigation of the scaffolds design more scientifically;(2)to build a novel biphasic scaffold with a gradient structure to meet the needs during the osteochondral repair process.To achieve the above goals,the main work of the paper is:(1)taking FCBPSS theory as the scaffold design guidance in the osteochondral reconstruction system;(2)investigating a proper material composition and formulation for scaffolds construction;(3)proposing a gradient structure scaffold with sufficient strength;(4)constructing the scaffold via 3D printing and testing the performance of the scaffold.Through the above work,the main conclusions are:(1)the investigation of the scaffold design can be proceeded more scientifically under the guidance of FCBPSS theory;(2)scaffolds with gelatin/hyaluronic acid(GelMA/HA)hydrogel as chondral phase and polylactic-coglycolic acid/poly-ethylene glycol(PLGA/PEG)composite as bony phase can be constructed by 3D printing and suitable for cell growth,proliferation and differentiation;(3)this paper proposed a reasonable scaffold design,and the scaffold with a gradient structure can meet the needs of the human microenvironment.The main contribution of this paper is:(1)taking the FCBPSS theory as the supportive design theory in scaffold investigation domain,which can provide theoretical guidance for the design of scaffold;(2)constructing a novel biphasic scaffold with gradient structure and making further exploration and contribution to the development of osteochondral tissue engineering.
