Currently,human glioma still is the higher incidence of all malignant intracranial tumor.Furthermore,mostly because of that it usually showed invasive growth,easy to relapse,and abundanted blood vessel,due to the surgery,radiation,chemical and genetic drug therapy are difficult to cure.The survival period of many high malignancy patients with glioma are 1 to 2 years.Therefore,the glioma seriously influence threaten the health of humanity.So,it is a difficult problem for all doctor of the neurosurgeons and tumors to utmost prolong survival time and to improve the life quality of patients with glioma.Up to now,people find that glioma is a polygenic disorder and the results of glioma development is mainly the activation of oncogene and tumor suppressor gene inactivation and produced a series of genetic abnormalities and the dynamic evolution,Therefore,although some important cancer biomarker were found,it is necessary to study that the more detailed pathological mechanism of that in glioma.Intercellular adhesion molecule-1 is a kind of immune globulin on cell of lymphocyte,monocytic,tumour,surface,which takes part in many physiological or pathological processes by mediating interactions between cells and cell and cells and extracellular matrix.It also as an important cellular adhesion molecules,which has a close relationship with many tumors of their genesis,development,metastasis.There were some evidences indicated that the role of ICAM-1 in central nerveous system was very important.Lately,some scholar find that the gliom's invasion,infiltration and so on have a close relationship with ICAM-1,especially Which may serve as indicators of invasion and malignant phenotype in the future.Matrix metalloproteinase(MMPs)are a family of zinc-dependent endopeptidases.Up to now,human fand exceed 20 kinds MMPs in all.They are grouped into collagenases,gelatinases,stromelysins and so on,which's group is based on domain substrate preference and that plays an important role in the degradation of extracellular matrix in various physiological and pathological conditions.More and more evidences have suggested that MMP-2 contribute to cancer cell invasion of the surrounding normal tissues especially in human gliomas,which make use of MMP-2 degradation,destruction of tumor cells in the ECM collagenase,and invasion nnormal and distant metastasis tissue along the basement,What's more,have a more close relation to human glioms.The lately research indicated that ICAM-1 MMP-2 maybe has emerged as an important cancer biomarker in gliom,But,Although they are all over expressed in glioma tissues,the detailed role and relationship system in gliom between ICAM-1 and MMP-2 is unknown now.So,In this study,the topic is to explore that theprotein expression and relevance of ICAM-I and MMP2 and The aim also is to To reveals the relationship of ICAM-1 and MMP-2 with glioma malignant degree and prognosis.Research methods56 Tumor materials and 10 with brain trauma hospitalized tissues were selected from the Department of Neurosurgery of the Fifth Hospital of Zhengzhou University as research objects and control group in 2012.4.30-2016.4.30.All patients which have a complete clinical data and had no history of chemo therapy,radio therapy or other treatment before.56 Tumor tissues which were comfirmed to be glioma tissues by pathologists.Using immunohisto chemistry and multivariate logistic method to detect ICAM-I and MMP-2 protein expressions and the relationship of ICAM-1 and MMP-2 with glioma malignant degree and prognosis in 56 glioma and 10 healthy brain tissue samples.ResultsThe expression of ICAM-1: 56 cases in human glioma and 10 cases in normal brain tissue of positive expression rate of ICAM-1 protein Was 67.86%(38/56)and20.0%(2/10).The expression of ICAM-I in glioma was significantly higher than healthy brain tissue(P<0.05).The positive rate of ICAM-I in high grade(WHO?-?)and low grade(WHO?-?)was respectively84.38%(27/32)and 45.83%(11/24)in human gliomas,The expression levels of ICAM-I protein were significantly higher in high grade than low grade of glioma groups(P< 0.05);In the contol group,low grade(WHO?-?),high grade(WHO?-?),the positive rate of ICAM-1 protein was gradually increased.(?2=9.340 P<0.05).The expression of MMP-2: 56 cases in human glioma and 10 cases in normal brain tissue of positive expression rate of MMP-2 protein Was73.21%(41/56)and10.00%(1/10).The expression of MMP-2 in glioma was significantly higher than healthy brain tissue(P<0.05).The positive rate of MMP-2 in high grade(WHO?-?)and low grade(WHO?-?)was respectively 90.63%(29/32),and 54.17%(13/24)in human gliomas,The expression levels of MMP-2 protein were significantly higher in high grade than low grade of glioma groups(P< 0.05);In the contol group,low grade(WHO?-?),high grade(WHO?-?),the positive rate of MMP-2 protein was gradually increased.(?2=9.722 P<0.05).The relevance between ICAM-I and MMP-2: The expression of ICAM-I and MMP-2 in glioma was 67.86%(38/56)and 73.21%(41/56)in human gliomas.the expression of ICAM-I was positive correlated with that of MMP2 by applicationing spearman rank correlation analysis(r =0.654,P<0.01).In Univariate analysis showed that patient age,the degree of tumor surgery resection,pathological grading,radio and chemo therapy after surgery,and The expression of ICAM-I and MMP-2 are meaningful to the postopterative 3-year survival rate of patients.Following Multivariate regression analysis found that degree of tumor surgery resection,chemo therapy after surgery,The expression of ICAM-I and MMP-2 were main prognositic factors influencing surival.Especially,the pathology classification directly affects the prognosis of glioma patients,The excess expression of ICAM-I and MMP-2in glioma,which have a bad affect for prognosis of glioma patients.Conclusions1.The expression levels of ICAM-I and MMP-2 were significantly higher in human glioma than in lealthl brain tissue.In the contol group,low grade(WHO?-?),high grade(WHO ?-?),the positive rate of MMP-2and ICAM-I protein was gradually increased.The result shows that The expressions of ICAM-I and MMP-2 are closely related to the malignant degree of gliomas.2.We found that both ICAM-I and MMP-2 expression were significantly higher in high grade than low grade of glioma groups.So,ICAM-1 Expression was probably a positive correlated with the expression of MMP-2.They may common contribute to the evolve of glioma.3.The pathological grading,degree of tumor surgery resection,chemotherapy,The expressions levels of ICAM-I and MMP-2 in human glioma,Observed in the clinical treatment of these indicators for judging theprognosis of glioma has more significance.
