Skeletal muscle belongs to striated muscle,which is a kind of muscle tissue in the animal body,and it is mainly distributed in the trunk and limbs.Skeletal muscle is a highly differentiated tissue,it has many physiological functions,such as maintaining body posture,generating body movement and metabolizing glucose.However,in the course of physical movement or under some pathological conditions,skeletal muscle easily to be damaged or arise metabolic disorders,and will affect the patient's normal action and life seriously.Accordingly,it is of great clinical significance to treat and repair damaged skeletal muscle,maintain normal metabolism of skeletal muscle and improve skeletal muscle function.Ginsenoside Rg1,a panaxitriol,is one of the main active components in ginseng.In recent years,the medicinal value of ginsenoside Rg1 has received extensive attention.Many studies have found that it has a lot of important effects on human health,such as anti-cancer,anti-radiation,anti-aging and so on.However,the ginsenoside Rg1 on the regulation of development and metabolism in skeletal muscle less reported.Therefore,this study mainly investigated the effects of ginsenoside Rg1 on skeletal muscle proliferation,differentiation and glucose uptake.[Experimental Objective] Analyze the effects of ginsenoside Rg1 on the proliferation,differentiation and glucose uptake of skeletal muscle myoblasts C2C12 in mouse,study the protective effect of ginsenoside Rg1 on skeletal muscle.[Experimental Method] Skeletal muscle myoblasts C2C12 in mouse were incubated with different concentrations of ginsenoside Rg1.Cell counting,MTT and Flow cytometry was used to investigate the effect of Rg1 on proliferation in C2C12 cells;Western blot was used to detect the expression of MyHC and MyoG,and investigated the effect of Rg1 on differentiation in C2C12 cells;Glucose-oxidase method was used to detect glucose consumption of C2C12 cells,Western blot was used to detect the expression and translocation of GLUT-4,and investigated the effect of Rg1 on glucose uptake in C2C12 cells.[Experimental Results] After the mouse skeletal muscle myoblasts C2C12 was incubated with a certain concentration of Rg1(10-4,10-3,10-2,10-1mM),the viability of C2C12 cells was effectively improved and their proliferation was boosted;At the same time,Rg1 up-regulated the expression of myosin heavy chain MyHC and myogenin MyoG,promoted differentiation in C2C12 cells;In addition,Rg1 increased the glucose consumption of C2C12 cells and promoted the expression and translocation of glucose transporter-4,which could promote the uptake of glucose in C2C12 cells.Based on the above experimental results: ginsenoside Rg1 can promote the proliferation,differentiation and glucose uptake in skeletal muscle myoblasts C2C12 in mouse.The results shows that ginsenoside Rg1 can participate in the regulation of skeletal muscle development,differentiation and metabolic.It provides a theoretical basis for the clinical research and application of ginsenoside Rg1 in the repair of skeletal muscle injury,the treatment of muscle atrophy and the improvement of skeletal muscle function.
