Effects Of Danshao Granular Ⅲ Inducing P-Akt Signal Transduction Pathway On The Treatment Of Henoch-schonlein Purpura Nephritis Rats

Objective:To investigate the mechanism underlying Danshao Granule III treating henoch-schonlein purpura nephritis(HSPN)rats.Methods:SD male rats were randomly divided into 5 groups,the control group,the model group and the Danshao Granule groups with low,middle and high doses respectively,5 rats in each group.Then,the Danshao Granule groups were treated with different doses of Danshao Granule III.The histopathological changes of the kidney tissue were observed.The renal glomerulus basement membrane and podocyte were observed with the transmission electron microscope.The 24h urinary protein quantity was determined with BCA method.The protein expression of p-Akt was detected by Western-blot.The mRNA expression of Akt was determined by RT-PCR.Results:The pathological results showed that renal injury was significantly observed in the model group,which was reserved by the administration of Danshao Granule.The renal glomerulus basement membrane was clear and complete,and the foot process structure was compact arrangement,abnormalities didn’t been yet found in the control group.But,the renal glomerulus basement membrane became folded,and the foot process fusion could be obviously observed in different degree,electron dense deposits could be found in mesangial area.But,the administration of Danshao Granule significantly improved the HSPN injured the renal glomerulus basement membrane and foot process fusion.The 24h urine protein quantity in the model group was higher than that in the control group(P<0.05),while that was lower in the Danshao Granule groups compared with the model group(P<0.05).The protein expression of p-Akt was higher in the control group than that in the model group(P<0.05),while that in the model group was lower in comparison with the Danshao Granule groups with middle and high doses(P<0.05).The mRNA expression of Akt were different in each group,but no significantstatistical difference was found between each two groups(p>0.05).Conclusion:Danshao Granular Ⅲ could effectively increase the generation of p-Akt proteins and maintain the effect of PI3K/Akt signaling pathways on inhibiting podocyte from cell fusion and apoptosis,thus to improve the symptoms of HSPN.
