Objective:To measure the effects of several traditional Chinese medicines on human colorectal cancer Lovo cells and investigate the inhibitory effect of atractylenolide II on Lovo cells and its mechanism.Methods:Lovo cells were cultured in vitro,respectively added different traditional Chinese medicines at different concentrations,then continued to incubate for 24h,the proliferation ability of Lovo cells were detected by MTT assay.The apoptosis of Lovo cells impacted by AT-? were measured by Flow cytometry(FCM),and the effect on the expression of PARP1 and caspase-3 were detected by the Western blot.Results:(1)Different concentrations of astragaloside and diosgenin solution had no obvious effect on the proliferation of Lovo cells,and its ability of proliferation was not correlated with the concentration of solution.Baicalin solution could promote the proliferation of Lovo cells to a certain extent,but its proliferation ability was not related to the concentration of baicalin.The effect of different concentrations of AT-? on the proliferation of Lovo cells was significantly higher than that in the control group.The inhibitory effect on the proliferation of Lovo cells was stronger with the increase of drug concentration.When the concentration was more than 300?g · ML-1,the inhibition rate was nearly 50%.(2)The expression of connexin 43(Cx43)in Lovo cells was not significantly affected by different concentrations of astragaloside,diosgenin,baicalin and AT-?I.(3)Low dose(50?g · mL-1),medium dose(150?g · mL-1)and high dose(300?g · mL-1)of AT-? can promote the apoptosis of Lovo cells,especially high dose of AT-?,and the difference was statistically significant(P<0.01).(4)AT-? 100?g · mL-1 began to shear PARP1 and caspase-3,with the increase of drug concentration,the effect was significantly enhanced,suggesting that AT-? can significantly change the expression of PARP1 and caspase-3,and play an important role in promoting the apoptosis of tumor cells.Conclusion:Only AT-? can inhibit the growth and proliferation of Lovo cells,regulate the expression of PARP1 and caspase-3,then promote the apoptosis of cells.AT-? can significantly inhibit the proliferation of Lovo cells and induce the apoptosis,its molecular mechanism may be related to the regulation of the expression of PARP1 and caspase-3.
