Objective:To explore the effects of Fructus Broussonetiae decoction which is from Bushengyijing Fang on the model of PC 12 cells induced by H2O2.Methords:The effects of viability of PC12 cells under different concentrations of fructus broussonetiae was detected by MTT,and so was the survival rates of PC 12 cells divided into the control group,model group and low-dose?medial-group and high-dose group.The apoptosis-related protein such as Bax?Bcl-2?CytochromeC and Caspase 3were determined by Western Blot analysis.Results:Fructus Brousonetiae decaton playerd a role in the protection of PC 12 cells in a doses-dependent manner?P<0.05?,and it could inhibit the apoptosis of PC12 cells cells induced by H2O2,and dawn-regulate the expression of cleaved-Caspase3,Bax,cytochromeC in a doses-dependent manner?P<0.05?.Conclusion:The Frutus Broussonetiae decotion was able to protect PC12 cells from apoptosis induced by H2O2,due to the effect of up-regulate the ratio of Bcl-2/Bax,the mitochondrial integrity,preventing the release of cytochrome C and dawn-regulate the expression of cleaved-Caspase3. |
