| Objective:To explore the effect of tendon-relaxing and blood-activating Chinese herbs on the homing of rat BMSCs in vivo.Method:Rat intervertebral disc degeneration models are established by anulus puncture in tail and tested by MRI.50 rat disc degeneration models are randomly divided into 5 groups:group A:rats are injected with bone marrow mesenchymal stem cells(BMSCs)and filled of high dosage Chinese herbs;group B:rats are injected with BMSCs and filled of medium dosage Chinese herbs;group C:rats are injected with BMSCs and filled of low dosage Chinese herbs;group D:models are injected with BMSCs and filled of pure water as the same volume as Chinese herbs;group E:models are injected with PBS as the same volume as BMSCs and filled same water.Rats models in group A?B?C and D,are tested by IVIS Image System at the 3rd day?1st week?2nd week and 4th week.Moreover,all models are examined by MRI and histopathology methods afer 4 weeks.Result:The disc degeneration standard is grade 3 before operation of herbs and BMSCs.Fluorescence luminance in group A?B?C and D,significantly differs at the 3rd day?1st week?2nd week and 4th week(P<0.05)and damps during experiment;the AR of the four goups does not occure significant difference at the 3rd day and 1st week(P>0.05),but differs significantly at 2nd week and 4th week(P<0.05).There are significantly different(P<0.05)in part of MRI and histopathology grading score in all groups.The content of proteoglycan within group A?B and E,has significant difference(P<0.05).The difference of collagen ? is also significant(P<0.05)within all groups.Conclusion:The tendon-relaxing and blood-activating Chinese herbs may protect the mumber and vitality of BMSCs during migration in vivo,and not increase the cell numbers.The new treatment strategy,herbs associated with stem transplant,can raise the content of proteoglycan and collagen ? in degenerative disc,and prevent disc from degenerating in some degree,so it will be a potential method in the future. |
