Objective:To investigate the role of ginsenoside Rbl and autophagy in order to find a kind of traditional Chinese medicine which can reduce brain injury after cerebral ischemia and hypoxia.Methods:Thirty rats were randomly divided into 6 groups(n=5),high dose group,middle dose group,low dose group,blank control group,sham operation group and normal control group.The rats in the high dose group and the middle dose group and the low dose group were injected intraperitoneally at 4 weeks after the model establishment at the concentration of 10mg/kg,5mg/kg and 2.5mg/kg respectively.For the sham operation group and chronic ischemia group,0.5ml of ultrapure water was given as the control.?At the same time,the water maze test was carried out for 45 days after modeling and 90 days after modeling.The experiment was conducted by positioning the navigation experiment and working memory experiment.By observing the learning and memory ability of rats to judge their cognitive ability.?The contents of Caspase-3 and LC3B in the brain tissue of rats were measured by Western blot to understand the level of autophagy in the brain tissue of rats(P<0.05)? Read the brain tissue(the frontal and hippocampus)after HE staining optical microscopy,observe(1)brain:cortical neurons with or without degeneration,necrosis,glial cells with or without proliferation,medullary edema.(2)hippocampus:hippocampus of the vertebral cells with or without degeneration,necrosis and other lesions.And the severity of the lesion was scored.Results:(1)Compared with the normal control group and the sham operation group,the learning and memory abilities were obtained after the initial maze of the sham operation group.The rats in the model group had obvious cognitive dysfunction after 45 days and 90 days.The duration and latency of the model group were significantly longer than those of the model group(P<0.05),and the distance between the two groups was significantly longer than that of the model group(P<0.001).The performance was more Significantly.(P<0.05),and the search distance(P<0.001)of the rats in the model rats was significantly shortened,Ginsenoside Rbl can decrease the duration and latency(P<0.05),and the higher the concentration was,the more obvious the effect was(P<0.05).The expression of Caspase-3 was decreased in all the cerebral ischemia and hypoxia model group(P<0.05),and the content of LC3B decreased(P<0.05),and the expression of Caspase-3 was decreased by ginsenoside Rbl While promoting the expression of LC3B,the higher the concentration the more obvious effect.? After the HE staining,the brain tissue of cerebral ischemia and hypoxia was observed.The structure of the brain was similar to that of the normal control group.Most of the cortical structures disappeared and the neuronal necrosis were localized.The local tissue was highly loose and showed vascular and glial cell proliferation.Hippocampus 1 and 2 area,the structure are unclear,most pyramidal cells necrosised,disappearance,residual individual structural unclear necrotic cells;the 3th area regions of the majority of pyramidal cell volume degeneration,nuclear condensation,irregular shape;lesion score shows that the patients in the high dose group and the middle dose group had mild brain tissue lesion,the low dose group was moderate brain lesion,the blank dose group was extremely severe brain tissue lesion,the sham operation group was mild brain tissue lesion.The Ginsenoside Rbl can improve the brain tissue necrosis in the model group and the higher the concentration,the more obvious the improvement effect.Conclusion:Ginsenoside Rbl can reduce the degree of brain injury in rats with hypoxia by improving the learning and memory ability of rats with cerebral ischemia and hypoxia,regulating the expression of autophagic factors and reducing the necrosis of brain tissue...
