| Objective:Biyuan Mixture is an in-hospital prescription of Jiangsu Province Hospital of Traditional Chinese Medicine and it is effective in the clinical treatment of acute and chronic sinusitis.The aim of this study is to analyze the mechanism of Biyuan Mixture by observing the effect of the symptoms and signs of acute sinusitis model before and after treatment,as well as the effect of P38MAPK signaling pathway,IL-9 and IL-10 on nasal sinus mucosa membrane.Sincerely hope that the experiment for the treatment of acute sinusitis to provide different ideas and objective basis for better use in clinical treatment.Methods:65 SD rats,male and female half,were randomly divided into blank group and model group,the blank group(no building dosing)have 5 rats,including 3 females and 2 males,as well as the model group have 60 rats,including 30 females and 30 males.The modle group is divided into traditional Chinese experimental group,western medicine control group and saline control group,there are 20 rats in each group,10 female rats and 10 male rats.The experimental group was treated with Biyuan Mixture,the western medicine control group was given a gavage of clarithromycin dispersible tablets and Eucalyptus lemon pinene enteric soft capsule,and the saline control group was taken gavage method with physiological saline.The rats in the model group were modeled by modified Hinni method.After the model was successfully established,the rats were treated by intragastric administration.The rats were sacrificed at the first day,1 week,2 weeks and 3 weeks(Recorded as 0d,7d,14d,2 1d)after the completion of the modeling.Dissection of nasal mucosa and using Western blotting method to detect the levels of p-p38,IL-9,IL-10 on the mucosa tissue membrane.Results:l.There was statistically significant difference(P<0.05)in the white blood cell count between the blank group and the model group(including the saline control group,western medicine control group and the traditional Chinese medicine experimental group),but the mean value of white blood cell count in the model group was within the normal range.2.After intragastric treatment,The symptom score of the western medicine control group was lower than that of the saline control group at 1 week after the model was completed,there was statistically significant difference between the two groups(P<0.05).In the experimental group,the symptom score began to be lower than that of the saline control group at 2 weeks after the completion of the model,the difference has statistically significance(P<0.05).Until 3 weeks after modling,the symptom scores of traditional Chinese medicine experimental group and western medicine control group were lower than saline control group,the difference was statistically significance(P<0.05).However,there was no significant statistically difference in the symptom score between the experimental group and the western medicine control group(P>0.05).3.There was no significant statistically difference of the p-p38 level between the Chinese medicine experimental group and the saline control group in group comparison(P>0.05)and significant statistically difference in the group comparison of western medicine control group(P<0.05).4.There was significant statistically difference of the IL-9 content between the Chinese medicine experimental group and the saline control group in group comparison(P<0.05)and no significant statistically difference in the group comparison of western medicine control group(P>0.05).There was no statistical difference between the experimental group and the saline control group(P>0.05).5.There was statistically difference of the IL-10 content between the Chinese medicine experimental group and the western medicine control group in group comparison(P<0.05)and no statistically difference in the group comparison of saline control group(P>0.05).Comparing Chinese medicine group with saline control group and western medicine control group,the difference has no statistically significance(P>0.05).Conclusion:Biyuan mixture can improve the symptoms of acute sinusitis in rats,and has similar effect with western medicine group.However,there was no significant effect of Biyuan Mixture on p-p38,IL-9 and IL-10 in nasal sinus mucosa. |
