Objective: To investigate the relationship between the expression of VEGFR2,HER2 and TP in gastric cancer and the degree of malignancy of gastric cancer, and the significance of combined detection the three markers in assessing the malignant degree of gastric cancer and its clinical value. Methods:Retrospective analysis the results of VEGFR2,HER2,TP and related clinicopathological data(age, gender, tumor size, tumor location, differentiation, depth of invasion,TNM stages, regional lymph node metastasis, distant metastasis, vascular invasion and nerve invasion)in 169 patients with gastric cancer which were treated In the Second Affiliated Hospital of Dalian Medical University gastrointestinal surgery from January 2015 to October 2016.Using SPSS17.0 software system for statistical analysis, the relationship between VEGFR2,HER2,TP expression level and clinicopathological data was analyzed by the Mann-Whitney test and the Kruskal-Wallis test. The correlation between the expression of VEGFR2,HER2,and TP was analyzed by Spearman rank correlation analysis.According to the results of statistical analysis to explore the above three markers of immunohistochemical expression of gastric cancer and the relationship between the degree of malignancy, and the combined detection of the three malignant degree of gastric cancer significance and clinical value. Results: The expression of VEGFR2 in gastric carcinoma was correlated with the depth of tumor invasion(p = 0.013),clinical TNM stage(p = 0.019),lymph node metastasis(p = 0.007)and vascular invasion(p = 0.002),and positively correlated with he depth of tumor invasion(r=0.217,p=0.005)and clinical TNM stages(r = 0.237,p = 0.002),but not with gender, age, tumor size, tumor location, differentiation and distant metastasis and nerve invasion(p>0.05).The high expression of HER2 was only correlated with the degree of differentiation(p=0.001)and negatively correlated(r =-0.286,p<0.001),but not with sex, age, tumor size, tumor location, depth of tumor invasion, clinical TNM stages and regional lymph node metastasis, distant metastasis and vascular and nerve invasion(p>0.05).The high expression of TP was correlated with the depth of invasion(p = 0.016),lymph node metastasis(p = 0.012),clinical TNM stages(p = 0.010)and distant metastasis(p = 0.019),and positively correlated with he depth of tumor invasion(r=0.214,p=0.005)and clinical TNM stages(r=0.196,p=0.011),but not with gender, age, tumor size, tumor location, differentiation degree and vascular and nerve invasion(p>0.05).VEGFR2 was positively correlated with TP(r=0.200,p =0.009).There was no significant correlation between HER2 and VEGFR2 and TP expression(p>0.05).Conclusion: The high expression of VEGFR2 is related to the depth of invasion,TNM stages, lymph node metastasis and vascular invasion. It may be an important index to evaluate the degree of malignancy of gastric cancer, and it is positively correlated with the malignancy of gastric cancer. The high expression of HER2 was negatively correlated with the differentiation degree of gastric cancer. The expression of HER2 was selective,HER2-positive gastric cancer may have a lower degree of malignancy. The high expression of TP may be related to the depth of invasion, lymph node metastasis,TNM stages and distant metastasis of gastric cancer. It may be one of the indexes to evaluate the degree of malignancy of gastric cancer, and it is positively correlated with the malignant degree of gastric cancer. The expression of VEGFR2 was positively correlated with the expression of TP,while the expression of HER2 had no significant correlation with the expression of VEGFR2 and TP.It is suggested that HER2 may not be directly involved in the angiogenesis of gastric cancer and not increase the malignancy of gastric cancer.VEGFR2,TP may have synergistic effect on the angiogenesis of gastric carcinoma and aggravate the malignant degree of gastric cancer. The combination detection of the three indexes is of great value for evaluating the malignancy of gastric cancer, and has guiding significance for the choice of clinical targeted drugs and chemotherapy drugs.
