ObjectiveThe purpose of this study is to detect the expression levels of COX-2 protein and VEGF protein in NPC tissues and normal mucosa tissues of nasopharynx.Analysis the potential relationship between the expression of these two proteins and the occurrence of NPC,the relationship between the two proteins in the occurrence and evolution of nasopharyngeal carcinoma and their clinical significance.Materials and MethodsSpecimens from patients who undergoing nasal endoscopic nasopharyngeal new pharyngeal biopsy in the First Affiliated Hospital of Zhengzhou University during February 2015 to December 2016,the selected patients were not received with radiotherapy,chemotherapy and non-steroidal drugs before admission,and the data were complete.Each patient took two parts of the organization,one nasopharyngeal neonatal Department,the second is the nasopharyngeal surface is not prominent,naked eye class normal parts.Two parts of the organization by our hospital pathology were diagnosed as nasopharyngeal carcinoma,nasopharyngeal normal mucosal tissue,to meet the conditions of patients in 42 cases.The nasopharyngeal normal mucosal tissue of each nasopharyngeal carcinoma was selected as the control group.All the specimens were was stored on the refrigerator at-80 ?after biopsy,and all patients were confirmed on the informed consent form.The experimental methods involved in this paper are tissue extraction of total protein,BCA method for quantitative protein and Western Blot.The expression of COX-2 protein and VEGF protein in the experimental group and the control group were detected by the above methods.Analysis all the data by using SPSS21.0 software.The data obtained by Western Blot method must be tested by normality and variance homogeneity.The data of normal distribution were expressed by mean ± standard deviation,paired t test,independent sample t test,Pearson correlation analysis,P<0.05 as a Statistically significant for judging differences.Results1.The results of Western Blotting display that the level of COX-2 gene protein expression in nasopharyngeal carcinoma was significantly higher than that in their matched normal mucosal(0.824±0.147 vs 0.201±0.082,respectively,t=8.301,P=0.000).2.The results of Western Blotting display that the level of VEGF gene protein expression in nasopharyngeal carcinoma was significantly higher than that in their matched normal mucosal(0.709±0.103 vs 0.368±0.098 respectively,t=5.343,P=0.001).3.The level of COX-2 gene protein expression in nasopharyngeal carcinoma with cervical lymph node metastasis was significantly higher than without lymph node metastasis(0.878±0.069 vs 0.751±0.052,respectively,t=3.310,P=0.011),the level of COX-2 gene protein expression in nasopharyngeal carcinoma in TNM stage ?-?was significantly higher than that in stage ?-?(0.882±0.064 vs 0.747±0.046,respectively,t=3.844,P=0.005),the level of COX-2 gene protein expression was not associated with age,gender(P >0.05).4.The level of VEGF gene protein expression in nasopharyngeal carcinoma with cervical lymph node metastasis was significantly higher than without lymph node metastasis(0.748±0.056 vs 0.656±0.036),respectively,t=2.604,P=0.031),the level of VEGF gene protein expression in nasopharyngeal carcinoma in TNM stage ?-was significantly higher than that in stage ? ?-?(0.758±0.038 vs 0.658±0.036,respectively,t=4.279,P=0.003),the level of VEGF gene protein expression was not associated with age,gender(P >0.05).5.There was a positive correlation between COX-2 protein and VEGF protein expression in nasopharyngeal carcinoma(r=0.808,P=0.005)by Pearson correlation analysis.Conclusions1.The up-regulation of COX-2 gene protein was related to the incidence nasopharyngeal carcinoma.The level of COX-2 gene protein expression was not associated with age,gender,was associated with cervical lymph node metastasis,TNM stage,with cervical lymph node metastasis was significantly higher than without lymph node metastasis,TNM stage ?-?was significantly higher than that in stage ?-?.The up-regulation of COX-2 gene protein was related to the development of nasopharyngeal carcinoma.2.The up-regulation of VEGF gene protein was related to the incidence nasopharyngeal carcinoma.The level of VEGF gene protein expression was not associated with age,gender,was associated with cervical lymph node metastasis,TNM stage,with cervical lymph node metastasis was significantly higher than without lymph node metastasis,TNM stage ?-?was significantly higher than that in stage ?-?.The up-regulation of VEGF gene protein was related to the development of nasopharyngeal carcinoma.3.There was a positive correlation between COX-2 protein and VEGF protein expression in nasopharyngeal carcinoma.Suggesting that COX-2 protein and VEGF protein in the occurrence and evolution of nasopharyngeal carcinoma have synergistic effect.
