Objective: To investigate the effects of CUEDC2 (CUE domain containing 2) in invasion and metastasis of CRC (colorectal cancer).Methods: Paraffin sections and clinical data of 153 patients with CRC who underwent operation in Chinese PLA General Hospital from December 2015 to May 2016 were collected. The expression of CUEDC2 was analyzed by immunohistochemistry and univariate and multivariate analyses were carried out to investigate the association of the expression of CUEDC2 with lymph nodes metastasis. Sixteen fresh colon cancer tissues were collected from the General Surgery Department of PLA General Hospital -eight cases with lymph node metastasis and eight cases with non-lymph node metastasis.The CRC cell lines SW480, SW620 and LOVO were cultured. Real-time PCR(Polymerase chain reaction) and WB (Western blot) were used to detect the expression of CUEDC2 in colon cancer tissues and CRC cell lines. Immunohistochemistry was used to detect the distribution and expression of CUEDC2 in colon cancer tissues.Results: The immunohistochemical score of CUEDC2 in cases with lymph nodes metastasis of CRC was significantly higher than that of patients without lymph node metastasis (7.41 ± 1.46 vs 4.61 ± 1.27,P<0. 05). Multivariate logistic regression analysis revealed that high immunohistochemical (?8) score of CUEDC2 was a independent risk factor for lymph nodes metastasis in CRC patients (OR: 4.777,95%CI:2.827-8.071,P < 0.05). The invasion and migration ability of SW480, SW620 and LOVO cells in CRC cell lines were significantly increased (P <0.05). Real-time PCR results showed that the expression of CUEDC2 mRNA in colon cancer was significantly higher than that in patients with non-lymph node metastasis and the difference was statistically significant (P < 0.05, n = 8); The expression of CUEDC2 mRNA increased in SW480, SW620 and LOVO and the expression of LOVO and SW620 was significantly higher than SW480 (P <0.05, n = 3). WB analysis showed that the expression of the CUEDC2 protein in colon cancer was higher than that in the lymph node metastasis group and was statistically significant; The expression of CUEDC2 protein in SW480, SW620 and LOVO increased and the expression of LOVO and SW620 was significantly higher than that of SW480 (P < 0.05, n = 3). The results of immunohistochemistry showed that CUEDC2 showed moderate intensity staining in the nucleus and cytoplasm of the colon cancer tissue, the expression of CUEDC2 in CRC tissues without lymph node metastasis showed low intensity staining.Conclusions: (1) High CUEDC2 immunohistochemical score (>8) is correlated with lymph nodes metastasis in CRC and CUEDC2 may promote the metastasis. (2)CUEDC2 is highly expressed in CRC tissues and CRC cell lines with high invasion and migration ability, and CUEDC2 may play a role in promoting invasion and metastasis in CRC nuclei and cytoplasm.
