Objective:this study is to investigate the effects of Quercetin on the stress response induced by cadmium in rats by regulating Nrf2-Keap1 pathway.Through the vivo study,the mechanism of Nrf2-Keapl is studied,which lays a foundation for the study of the reproductive toxicity of cadmium and the protective effect of Quercetin on oxidative damage.Method:through feeding groups,timing of gavaging treatment,model testing,4 weeks after removal of testis,testicular tissue by test of glutathione peroxidase(Gsh-Px),and catalase(CAT),super Qi matter peroxidase(SOD)antioxidant enzyme levels,testosterone secretion and antioxidant pathways of Nrt2-Keapl related factor 2(Nrf2),glutathione peroxidase(Gsh-Px)and heme oxygenase 1(HO-1),gamma glutamylcysteine synthetase(gamma-GCS),quinone oxidoreductase 1(NQOl)mRNA and protein expression,further determination of Quercetin as antioxidant effects on cadmium.Results:1.By gavaging rats after 5 days as a cycleon,the body weight of rats are measured,finds that CD group rats are obviously lower than that of the control group and Quercetin group,Quercetin group's weight is higher than the control group.The body weight of Quercetin treatment group is much obviously higher than the cadmium.2.Through the testis.The necessary anti oxidase test,analysis of test data showes that compares with the blank group,the contents of MDA increase significantly in CD group(P<0.01).MDA content of quercetin group is lower than that of the control group(p<0.01),However,MDA content of quercetin treatment group is lower than that of CD group(p<0.01).Manifold matter peroxidase(SOD)activity,catalase(CAT),glutathione peroxidase(Gsh-Px),CD group is obviously lower than the normal group,the difference is so significant(p<0.01),Quercetin treatment group's anti oxidase is higher than the CD group,,the difference is very obvious(p<0.01).The data indicates that Quercetin has a good effect on cadmium inducing body damage,which could enhance the antioxidant response of the body.3.By Elisa enzyme linkes immunosorbent assay showes that comparing with the blank group,CD group,testosterone content decreases significantly(p<0.01),Quercetin group testosterone content increases significantly comparing with control group(p<0.01),Quercetin treatment group,comparing with the CD group has significant difference,but lower than that of the control group(p<0.05).The results showes that Quercetin could inhibit the decrease of the amount of testosterone secreted by cadmium and reduce the damage to the body.4.Basing on the testicular tissue HE staining analysis finds that,comparing with the control group,with the existence of a large number of testicular tissue cadmium group seminiferous epithelium exfoliated germ cells,germ cells decrease in the number,the number of central elongates sperm cells is reducing the phenomenon of seminiferous tubules,Quercetin treatment group off part of seminiferous epithelial cell injury,endothelial cell injury is not significant,Quercetin can alleviate testicular injury inducing by cadmium in rats.5.By q-PCR technology and Western Blot technology to detect Nrf2 protein antioxidant pathways Nrf2,HO-1,NQO1,Gsh-Px,R-GCS,Keapl mRNA and protein levels,finds that cadmium group,Keapl mRNA and protein levels is higher than the control group(p<0.01),but Nrf2,HO-1,NQO1,Gsh-Px.Y-GCS mRNA and protein levels is obviously lower than the control group(p<0.01,p<0.05).For Quercetin treatment group,the expression level of Keapl is significantly lower than cadmium group(p<0.01),and the levels of Nrf2,HO-1,NQO1,Gsh-Px,gamma-GCS is higher than those of the control group(p<0.01).Conclusion:Cadmium could damage the spermatogenic function of testis in rats,reducing the ability of anti oxidative reaction of testis tissue,and cause obvious damage to the reproductive system.Quercetin can activate the antioxidant pathway of Nrf2-Keap1,regulate the expression level of antioxidant enzymes,and interferes with cadmium damage.It can be concluded that Quercetin plays an important role in the defense of endogenous oxidative reactions inducing by cadmium.
