Objective:This experiment aimed to study purgative activity and relieve fever function of compatibilities with Rhubarb and Fructus Aurantii Immaturus,Chinese goldthread rhizome,Moutan Cortex,peach seed and Kansui Radix stir-baked with vinegar,as well as to determine the contents of free and total anthraquinones among five different compatibilities of Rhubarb.HPLC sim?Ltaneous determination of 7anthraquinone derivatives for the compatibility of herb-pair in Rhubarb.A HPLC method that pharmacology,pharmacokinetic and ser?m pharmacochemistry,was established by pharmacokinetic experiment of five different compatibilities.These studies on compatibilityr?Le of herb-pair in Rhubarb reveal the specific pharmacodynamic material basis and mechanism,which provides scientific references for further study of TCM connotations.Methods: Comparing the effects about normal purgative activity,function of colorim etric of phenol red,charcoal ink prop?Lsion rate,and Na+-K+-ATPase activity of compatibility of pair herbs in Rhubarb by metabolic cage integral method,colorim etric method of phenol red,effect of charcoal ink prop?Lsion,and experiment of Na+-K+-ATPase activity.injected yeast and endotoxin to obsevse relieve fever effects.After 0.5h of administrated,took and compared the changes of tamperatures between7 h.HPLC sim?Ltaneous determination of 7 anthraquinones derivatives with an Dikma Dimonsil-C18 analytical col?mn(250mm × 4.6mm,5?m)by gradi-entelution of 0.1%(v/v)phosphoric acid in water and acetonitrile.The flow rate was 1m L·min-1,and detection wavelength was set at 256 nm.After0.5h copied the mold with endotoxin,rats were given a single i.g dose.Anthraquinone components in plasma and concentrations of aloe-emodin,rhein,emdin,chysophanol and physcion were measured with HPLC.Pharmacokinetics parameters were determined from the plasma concentration-time data with the DAS2.0 software package.Results:1.The purgative activity of Rhubarb were mostly changed by different compatibilities.Compared with the normal group,Rhubarb group,Rhubarb and Moutan Cortex group,Rhubarb and Persicae Semem group in 15 g · kg-1 were increased significantly on the effects about normal purgative activity,function of colorim etric of phenol red,charcoal ink prop?Lsion rate,and Na+-K+-ATPase activity.And compared with Rhubarb group,the purgative activity on Rhubarb and Moutan Cortex group and Rhubarb and Persicae Semem group were increased significantly,and Moutan Cortex for the most.2.Compared with the model-set group,relieve fever of pair herbs in Rhubarb were changed after compatibility,which Rhubarb group and Rhubarb and Aurantii Fructus Immaturus groups had a significant relieve fever effects about fever catched from yeast and endotoxin.And,compared with Rhubarb group,Rhubarb and Aurantii Fructus Immaturus groups had a significant relieve fever effects about fever catched from yeast and endotoxin.3.All the calibration curves showed good linearity(r ? 0.999)in a relatively wide concentration range.The %RSD of pair herbs from Rhubarb of7 anthraquiniones was less than 3.0%,and the recoveries were94.54%-103.1%.The the 7 anthraquiniones derivatives were changed after compatibility.Comparing with single Rhubarb,Rhubarb and Persicae Semem,the sennoside B,rhein and physcion in Rhubarb and Moutan Cortex,Rhubarb and Kansui Radix were all increased,the sennoside A and emodin in Rhubarb and Coptidis Rhizoma,Rhubarb and Aurantii Fructus Immaturus were decreased,especially the group of Rhubarb and Coptidis Rhizoma.The chrysophanol in Rhubarb and Coptidis Rhizoma was decreasedsignificantly and sharply.4.After i.g compatibility of pair herbs in Rhubarb in rats,aloe-emodin,rhein,emdin and chysophanol were detected in plasma except physcion and sennosides.Comparing with single Rhubarb,emodin increasing after competed of pair herbs.The T1/2 of rhein in Rhubarb and Coptidis Rhizoma,Rhubarb and Aurantii Fructus Immaturus were increased,and Rhubarb and Persicae Semem,Rhubarb and Moutan Cortex,Rhubarb and Kansui Radix were decreased.The Cmax(mg/L)of chrysophanol in Rhubarb and Persicae Semem,Rhubarb and Moutan Cortex,Rhubarb and Kansui Radix went increased.Conclusion: Study on the change of purgative activity and relieve fever function from pair herbs in Rhubarb,and the purgative activity,relieve fever function and the content of anthraquinones in vivo and in vitro in Rhubarb competed with pair-herb had mostly changed by different compatibilities,which affecting the effect of the phase to the phase.The content of anthraquinones in Rhubarb was changed by different compatibilities,and that may contribute to the characteristics of efficacy to Rhubarb.
