Effect Of RhEPO On Expression Of NR1 Subunit Of NMDA Receptor Of Hippocampal CA1 Area,Anxiety Behavior And Learning And Memory Capacity In The Adolescent Of Neonatal Rat With Hypoxic-ischemic Brain Damage

Abstract/Summary:
Objective: To research the effect of rh EPO on expression of NR1 subunit of NMDA receptor of hippocampal CA1 area,anxiety behavior and learning and memory capacity in the adolescent of neonatal rat with hypoxic-ischemic brain damage(HIBD).Methods: Adopting the method of improved Rice to make the HIBD animal models in mice.Let the rats rest 1 hour after the operation,then put them in anoxic tank(containing 8% oxygen and 8% nitrogen)2 hours continuously.by random number table,7-day-old Wistar rats were divided into control group(n=40),HIBD group(n=40)and HIBD+rh EPO group(n=40).The control group only free right common carotid artery,not ligation,without oxygen treatment.HIBD+rh EPO group injected rh EPO(5000 u/Kg)immediately,HIBD and control group injected saline solution with same amount.24 rats were selected in each group by random number table in 3 hour,12 hour,24 hour and 48 hour points,then took their brain tissue specimens and using immunohistochemical staining technique to detect the expression of NR1 subunits of NMDA receptor in rats hippocampal CA1 are.12 rats were selected in each group randomly in 35 day,49 day and 63 day to complete the adolescent anxiety behavior experimental study.4 rats were selected in each group to completed 35 day adolescent rats learning and memory ability of experimental research.Results: 1.The weight of three groups has no significant difference before the operation(P >0.05).After the operation the weight in HIBD group was significant difference compared with the control group and the treatment group there was differents(P < 0.05).2.NR1 positive cell number were increased on the right side of hippocampal CA1 area in HIBD group at 3h,compared with the control group and the treatment group there was differents(P < 0.05).At the time of 12 h,24h,48 the NR1 positive cell number of hippocampal CA1 area in HIBD group were higher than that of the treatment group and control group(P < 0.05).and the treatment group were higher than control group(P < 0.05).24 h time points had the highest NR1 positive cell number.48 h time points had the highest NR1 positive cell number.3.In elevated plus-maze,at P35,P49 and P63 the number of arm entry has no significant difference in three groups(P >0.05).No matter the ratio of open arm entry or the ratio of open arm residence time all has no difference between the control group and the treatment group(P >0.05).In HIBD group,no matter the ratio of open arm entry or the ratio of open arm residence time all has significant difference compared with the control group and the treatment group(P < 0.05).4.In open field test,at P35,P49 and P63 the horizontal motion total distance has no significant difference in three groups(P >0.05).No matter the number into central region or the motion time in area centralis all has no difference between the control group and the treatment group(P >0.05).In HIBD group,no matter the number into central region or the motion time in area centralis all has difference compared with the control group and the treatment group(P < 0.05).5.In Morris water-maze,at P35 the average escape latency of HIBD group was longer than the control group and the treatment group and has difference(P < 0.05).The average escape latency has significant difference between the control group and the treatment group(P < 0.05).So as the result of the wear ring index.Conclusion: 1.rh EPO can reduce expression of NR1 subunit of NMDA receptor of hippocampal CA1 area of neonatal rats with HIBD.2.The adolescent anxiety emotion of wistar rat with hypoxic ischemic brain damage will increase,and the exogenous rh EPO can make the anxiety behavior back to normal levels.3.The adolescent learning and memory ability of wistar rat with hypoxic ischemic brain damage will reduce,the exogenous rh EPO can reduce brain damage and improve learning and memory ability,but can not back to normal levels.
