Background and Objective Systemic lupus erythematosus(SLE)is an autoimmune disease involving multiple organs.Lupus Nephritis is one of the common clinical manifestations of lupus nephritis(LN),and its pathogenesis has not been fully elucidated.The expression of Toll like receptor 9(TLR9)and monocyte chemoattractant protein-1(MCP-1)in renal tissues of patients with LN was detected by immunohistochemical SP method,and the significance of the expression of LN was analyzed.The results provide a new basis for further elucidation of the significance of TLR9 signaling pathway in the development of the disease.Research subjects and Methods From January 2013 to July 2016,46 cases of LN patients in the First Affiliated Hospital of Medical University Of Anhui Department of Nephrology were chosen.There were 6 males and 40 females,aged 34±10 years old.12 patients with renal resection in Department of Urology,in the First Affiliated Hospital of Medical University Of Anhui from January 2016 to July 2016,were selected.There were 2 males and 10 females,aged 34±10 years old.There were no systemic diseases such as diabetes.There was no serious infection and Trauma history within 3 months.The renal tissue from the location of the renal cyst,which is far from the renal tumor,was confirmed as a normal control group by light microscopy.The expression of renal tissue was detected by immunohistochemical SP method?Image-Pro Plus 6 analysis software was used to analyze the image.SPSS 16 was used for statistical processing.Results The expression of TLR9 and MCP-1 in lupus group was significantly higher than that in control group(P<0.05).In lupus,the expression of type IV was the highest.Type V was less than type IV.Type III was the least.TLR9 and MCP-1 were mainly expressed in renal tubular interstitium and slightly expressed in glomerulus.The expression of MCP-1 in lupus group was consistent with TLR9.The tubulointerstitial TLR9 and MCP-1 in SLE group were positively correlated with 24 h urine protein,renal tubular pathology scores,and systemic lupus erythematosus disease activity index(SLEDAI)scores(P<0.05),but negatively correlated with serum C3(P<0.05).However,it had no significant correlation with serum creatinine and estimated glomerular filtration rate.Conclusions TLR9,MCP-1 in the LN group is mainly expressed in renal tubular interstitium and significantly higher than that in the control group.In particular,the intensity of expression in the tubular interstitium can objectively evaluate and predict the severity of lupus nephritis.TLR9/MCP-1 signaling pathway may be a new target for LN therapy.
