Object To detect the expression of Fra-1 in human hepatocellular carcinoma(HCC)and to explore the clinical significance of Fra-1 in HCC.Methods Immunohistochemical staining was used to investigate the expression of Fra-1 in 66 cases of HCC tissues and their corresponding peritumorous tissues;Real-time quantitative PCR(q RT-PCR)and Western blot were used to assess the relative expression levels of Fra-1 in 19 pairs of frozen HCC tissues and their corresponding peritumorous tissues.Results Immunohistochemical staining showed that Fra-1 was specifically expressed in the nuclei of HCC tissues and the cytoplasm of adjacent peritumoral tissues.The expression rate of Fra-1 in the 66 HCC tissues was 57.6%,which displayed in the adjacent tissues was 21.2%.The expression of Fra-1 in tumor cells was related to s AFP(serum alpha fetal protein)and vascular invasion,while not to the other clinicalpathological features,such as tumor capsule,TNM stage and Edmondson classification.The results of Western blot and q RT-PCR showed that the expression level of Fra-1 in HCC tissues was higher than their corresponding adjacent tissues(P(27)0.01);Kaplan-Meier survival analysis found that over-expressed Fra-1 was correlated with poor overall survival(OS)and disease-free survival(DFS).Multivariate analysis identified Fra-1 as an independent prognostic factor.Conclusion Fra-1 may be involved in the formation of s AFP and the progress of HCC,it is an independent prognostic factor.It may provide some new ideas and theoretical basis for the dignosis and treatment of HCC in the future.
