The Effect On Inflammatory Factors And Nogo-A In Experimental Autoimmune Encephalomyelitis Mice Treated With Shengjiang Powder

ObjectiveTo explore the anti-inflammatory and neuro-protective effect of Shengjiang powder on rats with autoimmune encephalomyelitis by observing its symptoms,pathological changes,and evaluating its inflammatory factors and Nogo-A levels.Meanwhile provide the experimental evidence for the treatment of multiple sclerosis.Methods32 female Wistar rats were divided into normal group,model group,hormone group and traditional Chinese medicine group.The EAE model were induced by antigen made from guinea pig spinal cord homogenate.Normal group and model group were treated with saline,while hormone group and Chinese medicine group were treated with prednisone and Shengjiang powder respectively.Treatment began on the day we induce the model.Weight,neuro-functional score were recoeded every day.After a 16-day-treatment,all rats were killed to collect the serum and brain spinal cord tissues.The content of IL-33 and IL-35 in serum was detected by ELISA.The volume of TLR-4 was detected by QPCR,the content of Nogo-A was detected by immune-histochemical staining,and the tissues were sectioned and stained with hematoxylin and eosin(HE)to reveal inflammatory infiltrates.Resultsneuro-functional score:the neuro-functional score of normal group was 0,and the scores of other three groups decreased at the beginning,then slowly recovery.The scores of the three groups were comparatively higher than the normal group,which indicated a statistical significance on the difference(P<0.05).On the other hand,the score of two treatment groups were lower than the model group,and their difference appeared to be statistically significant as well.Also,the scores between the two treatment groups and the model group were obviously lower,which showed a statistically significant difference(P<0.05).The score of traditional Chinese medicine group was lower than that of the hormone group(P<0.05).Incubation period:compared with the model group,the incubation period of the two treatment groups was much shorter,which showed a statistically significant difference(P<0.05).On the other side,the incubation period of traditional Chinese medicine group appeared to be obviously shorter than the hormone group as well(P<0.05).Peak period:however,The difference between the three groups showed no statistical significance(P<0.05).Weight:the weight of the normal group grew continuely,but the ones of other three groups decreased in the early stage,then rise in the later stage.Weight loss:compared with the model group,there was no significant difference in weight loss between the two groups(P>0.05).But the weight loss appeared to be much lower in traditional Chinese medicine group than that of the model group,which difference showed a statistical significance(P<0.05).IL-33:compared with the normal group,the IL-33 content in rats of the three groups were significantly higher,which difference showed a statistical significance(P<0.05).Besides,each content of the two treatment groups was lower than the model group respectively,which difference were statistically significant(P<0.05).However,the content of the two treatment groups showed no statistical difference.IL-35:Compared with the model group,the IL-35 content of the two treatment groups were both higher,which was statistically different(P<0.05).However,we found no significant difference when made a comparison between the two treatment groups.TLR-4:compared with the normal group,the TLR-4 level of the three EAE groups were significantly higher,and the difference was statistically significant(P<0.05).Both the TLR-4 level of the two treatment groups were statistically lower than the model group,which were significantly different(P<0.05).However,those data of the treatment groups did not differ too much in statistic.Nogo-A:compared with the normal group,the Nogo-A level in rats of the other three groups were significantly higher,which difference was significant in statistic(P<0,05).We found the content of the two treatment groups were both lower than the model group,which difference appeared to be statistically significant(P<0.05).However,the two treatment groups showed no significant difference on Nogo-A level.HE staining:there were different degrees of inflammatory cell infiltration and demyelination on the section of EAE groups.The HE pathological scores of the three groups was obviously higher when compared with the normal group respectively,which showed a significant difference in statistic(P<0.05).On the other sides,the HE scores of the two treatment groups were much lower than the model group,which differences were both statistically significant(P<0.05).However,we found no statistically significant difference between the two treatment groups.ConclusionShengjiang powder can postpone the onset of EAE and alleviate the severity of the disease,meanwhile reduce the content of IL-33 in serum,EAE in IL-33 rats,restrain the expression of TLR4 and Nogo-A in nerve tissue,enhance the content of IL-35 in serum.All above suggest that the Shengjiang powder can reduce the inflammatory reaction of EAE and the damage of central nervous system,and has a potentially curative effect on EAE.
