Objective:Gastric cancer is one of the most common malignant tumors in China.Compared with advanced carcinoma of stomach,the patients who suffer from an early stage of gastric cancer had an exact clnical effect and higher five year survival rate.Previous research had suggested that the unregulation expression level of IncRNA in magligant tumors had positive correlation of cell cancerous cell biological behaviors.In order to improve the early diagnosis of gastric cancer,clinical and scientific research had been developed.Methods:1.Based on gastric adenocarcinoma?GA?related IncRNA expression profiling data,select IncRNA NR026797 as research object.qPCR was used to measure the expression level of IncRNA NR 026797 in 80 cases of GA and 30 cases of gastritis tissues.Stasitic analysis was applied to explor the relationship of IncRNA NR026797 and patients clinical features.2.Verification the expression level of IncRNA NR026797 in SGC7901,AGS,BGC-823,MGC-803 cell lines.SiRNA transfected cell lines to knock down the expression level of lncRNAN NR026797,then CCK-8 cell proliferation assay,Colony forming assay,Transwell migration and invasion assay were applied to detect the alteration biological behavior of GA cell lines.3.Estimating the expression level of MMP19 in GA tissues,analysing the relationship of MMP19 with clinical features and IncRNA NR026797.Results:1.Gene chips of high flux assay reveals that IncRNA NR026797 is a up-regulated lncRNA,its fold change is 5.7 compared to the paired adjacent non-neoplastic tissues.The results of qPCR showed that the up-regulated rate of lncRNA NR026797 in 80 cases of GA tissues is 66.25%,fold change is 8.05.Stasitic analysis its expression level with clinical features showed that it's significantly associate with tumor size?3cm?P=0.046?,poor differentiation?P=0.0028?,T4 stage?P=0.033?,Ki67positive?P=0.011?.2.Compared with GES-1,IncRNA NR026797 is over expression in GA cell lines?SGC7901,AGS,BGC-823,MGC-803?,which is the same as IncRNA expression profiling data and clinical GA tissues.Silencing IncRNA NR026797,significantly suppressing the proliferation?P<0.05?,migration?P<0.05?and invasion?P<0.05?in GA cells.3.MMP19 was over expressed in GA tissues,its fold change is7.25,up-regulated rate was 63.5%,compared with non-cancerous gastric tissues.qPCR results revealed that the MMP19 expression levels were positively correlated with tumor size,TNM staging and lymphnode metastasis.The expression of MMP19 were relevanced with the expression of LncRNA NR026797 with a correlation coefficient of 0.557?P<0.001?.ROC curve revealed that both MMP19 and lncRNA NR 026797 have a diagnostic value of GA.Conclusion:1.LncRNA NR026797 was over-expreesion in GA,this abnormal high-expression of lncRNA NR026797 may indicate patients suffer a lager tumor,poor differentiation,late TNM stageing GA,leading to poor prognosis.2.LncRNA NR026797 was over-expresion in GA cell lines,silencing the expression of IncRNA NR026797,the behavior of proliferation,migration and invasion in GA cell were significantly suppressed.3.MMP19 was high-exprssion in GA tissues,the up-regulated state was correlated with tumor size,TNM staging and lymphnode metastasis.The expression level of MMP19 had a positive correlation with IncRNA NR026797,further research is needed to explor the relationship between MMP19 and lncRNA NR026797.
