Objective:to investigate the expression and significance of New York-esop Hageal-1(NY-ESO-1)? melanoma antigen gene-As(MAGE-As)in breast cancer and analyze the difference of expression of NY-ESO-1 and MAGE-Asin different molecularsubtypes of breast caner and their correlation with cytotoxic T lymphocytes(CTL).Method:1.we collected 150 tissue specimensdiagnosed with breast invasive cancer in the breast surgery department from January 2014 to January 2016,and selected 50 cases with pathology diagnosed as normal tissue specimens form the 150 cases by random number method.2.Collected and organized the clinicopathological date of selected patients,including age,tumor size,axillary lymph node metastasis,pathologic stage,tumor thrombus,WHO histological grade.3.Divided the breast cancer specimens into Luminal A type ?Luminal B type ?HER-2 over expression type and Basal-like type according to St.Gellen classification criteria for breast cancer in 2013.3.Immunohistochemistry was applied to detect the expression of NY-ESO-1?MAGE-As and CTL in breast cancer tissues and in normal breast tissues.5.we analyzed the expression of NY-ESO-1 and MAGE-As in different molecular subtypes of breast cancer and their relationship with the infiltration degree of cytotoxic T lymphocytes.Results:1.There was no NY-ESO-1 and MAGE-As protein expression in normal breast tissue.The expression rate of NY-ESO-1 and MAGE-As in breast cancer was 8.7%(13/150)?38.7%(58/150),with significant difference in breast cancer tissue and normal breast tissue(P>0.05);There was no relationship between MAGE-As and NY-ESO-1(r=0.047 P= 0.565).2.There was only significant differences between NY-ESO-1 with histological grade(?2=16.66 P=0.000),and were no significant differences between NY-ESO-1 and the age of patients(?2=0.025 P=0.426)?tumor size(?2=1.353 P=0.245)?metastasis of axillary(?2=0.604 P=0.739)?vascular invasion(?2=1.001 P=0.159)?pTNM stage(?2=0.027 P=0.870).The expression of MAGE-As protein was no correlated with vascular invasion(?2=0.695 P=0.278)?the age of patients(?2=2.1 P=0.102)?tumor size(?2=1.875 P=0.171)?metastasis of axillary(?2=1.060 P=0.589)? histological grade(?2=4.243 P=0.120)? pTNM stage(?2=6.857 P=0.077)? ER stage(?2=2.513P=0.131)?PR stage(?2=1.949P=0.163).3.The expression of NY-ESO-1 and MAGE-As were significant difference in different molecular subtypes of breast cancer,and the positive rate of NY-ESO-1 was higher in TNBC compared to non-TNBC(P<0.05).there was no significant difference of CTL in different molecular subtypes of breast cancer(?2=8.990 P=0.438).4.The expression of NY-ESO-1 was correlated with the number of CTL(?2=8.917P=0.03)in the tissue of breast cancer,and NY-ESO-1 expression was significantly correlated with a higher level of CTL in TNBC(?2=5.593 P=0.018);there was no correlation between MAGEAs expression and CTL in different molecular subtypes of breast cancer excepted in HER-2 over expression type breast cancer(?2=5.818 P=0.016).Conclusion:1.NY-ESO-1 and MAGE-As protein are tumor-associated antigens with highly expression in breast cancer.2.The expression of NY-ESO-1 protein was correlated with histological grade and was no correlated with the age of patients ?tumor size ?metastasis of axillary ?vascular invasion?pTNM stage.The expression of MAGE-As protein was no correlated with the clinicopathological of breast cancer,including vascular invasion ?the age of patiens ?tumor size ? metastasis of axillary ? histological grade ? pTNM stage.3.NY-ESO-1 was specifically expressedand positively correlated with CTL in triple negative breast cancer;the MAGE-As protein was correlated with different molecular subtypes of breast cancer,and negatively correlated with CTL in HER-2 over expression type breast cancer.
