Purpose and Meanings:Luteolin,a component of the prescription of "Zhou Shi Ke Jin Yan Fang" proposed by the domestic scholar Zhou Zhongying,has the effect of inhibiting the growth of human lung cancer cells.Its mechanism of action also has been revealed.That offered a new direction for the study of tumor drugs.Methodology:The human lung cancer cell line H460 was cultured in vitro.The luteolin was diluted respectively into 10?M,15?M,20?M,25?M and 30?M solution.The growth status of lung cancer cells was examined by CCK8 after giving luteolin.The cell cycle was examined by flow cytometry as well as apoptosis.Detection of lung cancer gene expression change by PCR.the expression of cycle related proteins DUSP6,DLC-1,JNK1,P-JNK1,CYD1P1,EGFR,ERBB2,PAK1,C-jun,NCK1,MEKK1 were detected by Westemblot.Results:The results of CCK8 showed that luteolin could effectively inhibit the proliferation of human lung cancer cells,the degree of hinder was related to concentration and time,and the effect of luteolin solution on H460 cells at 10?M,15?M,20?M,25?M and 30?M respectively,after 24h,36h and 48h,the OD value was significantly different from that of the control group.Flow cytometry analysis showed that:The human lung cancer cells were treated with luteolin for 36hours,the proportion of cells in S phase increased,the proportion of the cell in G1 and G2 phase reduced.After contact with luteolin for 24 hours,the proportion of cells in S phase increased.Indicated that luteolin play a role in the arrest the cancer cell in S phase,which was dose-dependent.Annexin V assay results showed the proportion of of H460 cells increased in the Q2 period(late apoptosis)after treated with luteolin for 24h,,after human lung cancer cells H460 treated with luteolin for 36h,the proportion of cells in Q3(early apoptosis)ratio increases,which was concentration-dependent manner.H460 cells treated with luteolin for 36h,Results of lung cancer gene by PCR,CYP1B1,DLC-1,EGFR,DUSP6,ERBB2 expression well.Westernblot display DUSP6,DLC-1,P-JNK1 expression increased,CYD1P1,EGFR,ERBB2,NCK1,MEKK1,PAK1 expression decreased,and also were dose-dependent.JNK1 and C-JUN expression did not change significantlyConclusion:Lu can significantly inhibit the proliferation of human lung cancer cell H460 and block the cell cycle in S and G1 phase,which inhibits cell proliferation.The mechanism may be closely related to the activation of JNK pathway.
