Objective: To investigate the expressions of M-CSF,TNF-?, IFN-? and IL-10 in the giant spleen of liver cirrhosis in rats and its significance by establishing a model of liver cirrhosis in rats.Methods: Totally 62 healthy male SD rats were assigned to model group(n=50) and control group(n=12) randomly. Rats in model group were made to be liver cirrhosis and hypersplenism with 40% solution carbon tetrachloride in peanut oil and liquor. Rats in control group were given by gavage with normal saline in the same time. And the expressions of M-CSF,TNF-?,IFN-y and IL-10 in the spleen of the two groups were tested by the immunohistochemistry, Western-Blot method and RT-PCR. And we assessed the difference of the expression between the two groups by SPSS software.Results: The positive rates of M-CSF,TNF-?, IFN-y and IL-10 expression in model group were 60.71%?78.57%?64.29%and 87.71% respectively, Which in control group were 25%?33.33%?16.67%and 50% respectively. There was a significant difference between the two groups(P<0.05). The protein relative strength of M-CSF,TNF-?, IFN-?and IL-10 expression in model group were 0.63±0.58, 1.06±0.49, 0.99±0.38 and 1.12±0.42 respectively, Which in control group were 0.18±0.12, 0.52±0.27, 0.38±0.28 and 0.60±0.32 respectively. There was a significant difference between the two groups(P<0.05). The relative level of M-CSF,TNF-?,IFN-? and IL-10 mRNA in model group with liver cirrhosis were 2.06±0.11, 4.07±0.19, 2.98±0.11, 7.94±0.27 respectively, Which in control group were 1.01±0.05, 1.06±0.11, 1.00±0.31, 1.02±0.08 respectively. There was a significant difference between the two groups(P< 0.05).Conclusions: The expressions of M-CSF, TNF-?,IFN-? and IL-10 in giant spleen of rats with liver cirrhosis and hypersplenism increased abnormally. The ratio and regulation of pro- and anti-inflammatory cytokines were dysfunction. And that maybe play an important role in the development of peripheral cytopenias concurrent hypersplenism induced by cirrhosis and portal hypertension.
