Objective: In this paper, Haishefengshi capsules were used as research object to study its anti-rheumatic effect and the mechanism of action by establishing non-specific inflammatory model, pain model and immune inflammation model,which provides technical supports of pharmacological aspects for Haishefengshi capsules using to treat rheumatic diseases.Methods: Xylene-induced mouse auricle inflammation swelling test, mouse capillary permeability test, mouse granuloma formation experiment were used as anti-inflammatory model. Hot plate method analgesic experiment, heat water decoying analgesia test, formalin-induced pain test, glacial acetic acid-induced wrinkle method analgesia test were used to establish the analgesic effect model and mouse delayed immune response test was used as the immune regulation of the model.They all used to study Haishefengshi capsules in the treatment of rheumatism effect.By modeling Freund's complete adjuvant arthritis in rats, the content of PGE2 in the inflammatory tissue, serum SOD activity, MDA content and cytokine IL-1?, TNF-a content, mouse macrophage phagocytosis and mouse spleen lymphocyte proliferation can be measured to study the anti-rheumatic mechanism of Haishefengshi capsules.Results: Through the above pharmacodynamic study, it was found that haishefengshi capsules group had significant inhibitory effect on the swelling of the ear auricle inflammation, and the swelling rate of the high dose auricle inflammation was 64.33%. Application of Haishefengshi capsules could significantly reduce the permeability of mouse capillaries. There was a significant difference in the absorbance values between the high dose group and the model group(P<0.01), and the highest inflammatory exudation inhibition rate of the highest was 80.65%. The inhibitory rate of granuloma formation in the Haishefengshi capsules group were increasing and the highest inhibitory rate was 64.33%, which indicated that Haishefengshi capsules had significant anti-inflammatory effect on acute inflammation and chronic proliferative inflammation. In the hot plate method,Haishefengshi capsules significantly increased the normal pain threshold level(P<0.01). In the hot water tail flick test, the pain thresholds of the mice were significantly improved at 30 min and 60 min after the administration of the Haishefengshi capsules (P<0.01). The inhibitory effect of the mice gavage administration of Haishefengshi capsules on formalin-induced pain was significant(P<0.01) and high dose group had the lowest pain score (100.60±14.58). In the torture analgesia experiment,the high,medium and low doses of Haishefengshi capsules had different inhibitory effects on the writhing response of mice, and the high dose group showed more prominent on writhing frequency inhibition rate that was 48.89 %,indicating that Haishefengshi capsules have good analgesic effect, and certain continuity analgesic effect. The results of late-onset immunoreactivity in mice showed that the thickness difference of right hind foot pad before and after high-dose group was significantly different from that of model group (P<0.01). The anti-inflammatory analgesic mechanism experiment results showed that Haishefengshi capsules could decrease the content of PGE2 in the inflammatory reaction, increase the activity of serum SOD and decrease the content of MDA, and the effect of the middle and high dose groups was significant (P<0.05). In the macrophage phagocytosis experiments,the mice gavage administration of Haishefengshi capsules at low, medium and high doses can significantly improve the phagocytic function of phagocytic cells (P<0.01);immune regulation mechanism experiment found that the levels of IL-1? and TNF-?(P<0.05) were significantly decreased in the group of Haishefengshi capsules and the proliferation of splenic lymphocytes was regulated.Conclusion: Haishefengshi capsules have obvious anti-inflammatory, analgesic and immune regulation of the pharmacodynamic effect. The study of anti-rheumatic mechanism of Haishefengshi capsule show that Haishefengshi capsules are indirectly can indirectly regulate the body's free radical level by improving the activity of SOD and reducing the level of MDA to achieve anti-inflammatory analgesic effect. They can also promote the phagocytic effect of macrophages in order to achieve anti-inflammatory analgesic effect and promote the body damage repair. Other else,they can regulate the mouse spleen lymphocyte proliferation bidirectionally as well as the levels of cytokines IL-1? and TNF-a.
