Objective: Correlation of ADAM33 V4 gene polymorphism,VitD levels with asthma of children in Urumqi to provide guiding prevention and treatment of children with asthma in genetic level.Method: Select the same period in asthmatic children 5-15 years old living in Urumqi region of 154 people(Han 111,Uygur 43),while the establishment of a control group of healthy non-asthmatic children 120(Han 64,Uygur56),first using the polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP)to analyze ADAM33 V4 gene single nucleotide polymorphism(SNP),ELISA measure the VitD levels,and further stratified according Ethnic groups were performed V4 sites,VitD analysis and correlation analysis.Result:(1)V4 sites of the frequency distribution of three genotypes between cases and control groups were statistically significant differences(x2 =6.806,P=0.033),among the different ethnic groups showed no significant difference(test group : x2 =0.714,P=0.699;control group:x2 =5.898,P=0.052).(2)Vit D Levels in the case group and the control group values are different,the difference was statistically significant(t=2.341,P=0.020).(3)The correlation analysis between V4 sites of ADAM33 gene and the Vit D Levels.Conclusions:(1)ADAM33 V4 gene loci and asthma morbidity in children in Urumqi region correlated,but there was no difference among peoples.V4 position after the G point mutation may be a risk factor of bronchial asthma.(2)VitD level of morbidity in children with asthma correlated.(3)ADAM33 V4 does not affects the VitD level. |
