| Object:To investigate the effects and preliminary mechanism DiXiangZhi ZhuXiaoKe Fang which is used for treatment of diabetes in the Yi nationality in type 2diabetic rats.Methods:Adaptive feed after 1 week,60 male healthy adult Wistar rats were randomly divided into normal group and model group.The normal group is fed with normal diet and the model group is fed with high fat diet.After feeding for 4 weeks,the rats in the model group are fasted for at least 12 hours,and then injected intraperitoneally with streptozotocin(30mg/kg).Blood glucose is measured after 3 days and rats that did not develop diabetes continued to give additional STZ according to the aforementioned dosages and methods.All diabetic rats were randomly divided into normal model group,positive group,Dixiangzhizhuxiaokefang low dose and high dose The normal group continued normal feeding.Normal model group was given normal model 1mL/100 g distilled water by intragastric administration every day.The positive group was given 1mL/100 g saxaglitin aqueous solution by intragastric administration every day(dose 0.5mg/kg).Dixiangzhizhuxiaokefang low dose and high dose group were given 1mL/100 g corresponding dose of the drugs by intragastric administration every day(Dose of 0.4 g / kg in low dose group and 2.0 g/kg in high dose group).Rat body weight and blood glucose were measured weekly.After 5 weeks of treatment,fasting insulin and glycosylated hemoglobin were measured by ELISA kit.After 30 minutes of intragastric administration of 2g/kg glucose,total GLP-1 and DPP-4 levels in serum were determined by ELISA kit.After the rats were sacrificed,take the small intestine and carry out pathological examination.Results: 1.Results of model buildingAfter the injection of streptozotocin(STZ)in the model group,the fasting blood glucose and the random blood glucose reached the standard of mode and the probability of the model was about 85%.Normal group and model group showed an upward trend in body weight,fasting blood glucose in the model group increased rapidly from the second week to the third week.2.Changes in body weight and blood glucose after administrationAfter administration,the body weight of the normal group continued to grow;the rest of the body weight decreased but decreased slightly,and rebounded slightly in the fifth week.Compared with model group,the fasting blood glucose of positive group and DXZZ-H decreased gradually from the second week after treatment,and the difference was statistically significant at the 5th week(P<0.05).Compared with model group,the random blood sugar of positive group and DXZZ-H decreased gradually from the third week after treatment,and the difference was statistically significant at the 5th week(P<0.05).3.Effects of fasting insulin and glycosylated hemoglobin content after administrationThe fasting insulin content in model group was significantly lower than that in normal group(P<0.01);The fasting insulin content in the positive group and DXZZ-H group was higher than that in model group(P<0.05).The levels of glycosylated hemoglobin in model group were higher than those in normal group(P<0.05);Positive group and DXZZ-L group were lower than model group in DXZZ-H group,but there was no significant difference among the groups(P>0.05).4.Effects of administration on serum GLP-1 and DPP-4 levels after administrationCompared with the normal group,the total serum GLP-1 content in model group was not statistically significant(P>0.05);The total GLP-1 levels in the positive group and DXZZ-H group were higher than those in model group(P<0.05).There was no significant difference in serum DPP-4 level among the groups(P>0.05).5.Pathological examination of small intestineIn the normal group,positive group and DXZZ-H groups,the intestinal mucosa structure was complete,the model group was slightly damaged.In the model and DXZZ-L group,the intestinal villi had erosions,swelling and shedding,and the epithelial cells were sparse,disorder and come off,the telangiectasia of the lamina propria and inflammatory cells was observed.The positive group and DXZZ-H group did not have this phenomenon,but it was slightly shorter and atrophy,and the intestinal glands of the DXZZ-H group increased slightly.In the ileal L cells immunohistochemical detection,we found that the normal group、the model and DXZZ-L group L cells without significant proliferation,positive group was hyperplasia,the DXZZ-H group significantly hyperplasia.Conclusions:DangXiangZhizhuXiaoKeFang maybe protect the intestinal mucosa lesion caused by type 2 diabetes mellitus,promote the proliferation of L cells,increase the level of GLP-1 in rats,promote insulin secretion,reduce the blood glucose in type 2 diabetic rats,eventually play the role of anti-type 2 diabetes. |
