Rheumatoid arthritis(RA)is a systematic autoimmune disease,characterized by high disability rate.As a crucial reason for adult disability,the morbidity in China is up to 0.4%.Due to the pathogenesis is still unclear,there are no miracle drug for treating RA thoroughly in clinical.Drugs used for relieving syndrome posses obvious side effects.TCM was regarded as an crucial resource to discover natural medicine with low toxicity and good activity.In traditional Chinese theory,There are also many TCM with anti-rheumatoid arthritis activity such as Tripterygium wilfordii.Several TCM resourced from Vitex(Verbenaceae)have significant anti-inflammatory activity,including mature fruits of Vitex trifolia L.and Vitex negundo L.Our research group have found kinds of compounds possessing anti-inflammatory activity and uncovered its potiential mechanism based on research of Vitex plants for many years.Vitex.pierreana,an arbor of Vitex,which is recorded in “Flora of Li” used for treating liver disease and inflammatory disease by Li people.Fruits of Vitex plants which were subjected more attention were used as medicine materials.Because of scarity of Vitex medicine resource,we aimed to found more compounds possessing good pharmacological activity in leaves,thereby expanding Vitex medical parts.1.Study on anti-rheumatoid arthritis activity of V.pierreana: we studied anti-rheumatoid arthritis effect of 80% ethanol extracts of the leaves of V.pierreana on established Collagen induced arthritis model(CIA).The results showed that V.pierreana extracts could obviously improve swelling degree of rats paws,reduce arthritis scores,down-regulate pro-inflammatory cytokine IL-1?,IL-6,TNF-a and up-regulate anti-inflammatory cytokine IL-10.All evidence above could suggest V.pierreana have anti-rheumatoid arthritis effect.2.Qualitative and quantitative analysis of the components of V.pierreana :Qualitative analysis of V.pierreana was carried out by HPLC-MS,and matched with data base of Vitex compounds established by ourselves afford 7 known components:5-hydroxy-3,6,7-trimethoxy kaempferol(1),isovitexin(2),orientin(3),luteolin-3-O-glucoside(4),agnuside(5),2'-O-p-hydroxybenzoyl-8-epiloganic acid(6),vitexioid(7).Quantitive analysis of two main components isovitexin(2)and agnuside(5)were performed on HPLC,the contents of the two components respectively are 1.276mg/g crude drugs,0.1964mg/g crude drugs.3.Study on chemical components of V.pierreana : Phytochemical study on petroleum ether position and ethyl acetate position of V.pierreana leaves and branches extracts afford 18 compounds by TLC,silical gel column chromatography,LH-20 column chromatography and PHPLC,and then elucidated their structure by IR,UV,NMR,X-Ray.18 compounds were isolated among which 3 were novel sesquiterpenoids.Information of isolates were showed as follow(new compounds were marked by *):...
