Background:With the huge development of our economy,people’s living and medical level has been greatly improved,the World Health Organization released the "2016 World Health Statistics" report shows that China’s average life expectancy of 76.1 years.At the same time,the incidence of cancer also increased.China is a high incidence of esophageal cancer,morbidity and mortality rank first in the world,has caused a huge social burden.According to the annual report of China’s cancer status statistics,in 2011,about 218,957 people died of esophageal cancer,was classified as the fourth largest cause of cancer death [1].The results of a systematic review and meta-analysis of the influencing factors of esophageal cancer in Chinese population show that the incidence of esophageal cancer in China is mainly dietary factors such as eating dry and hard,mildew and so on.Smoking is also a related factor [2] Gene susceptibility may also be one of the causes of the disease.Of the various pathologic types,more than 90% are squamous cell carcinoma,the rest are adenocarcinoma or other special types.At present,surgery and radiotherapy and chemotherapy is still the main means of treatment of esophageal cancer,but in view of the current incidence of esophageal cancer increased year by year,while the early diagnosis of esophageal cancer is still low,advanced and can not be treated esophageal cancer treatment effect is poor,poor prognosis,Therefore,comprehensive treatment of patients with esophageal cancer to improve the prognosis of the main treatment model,including preoperative neoadjuvant,postoperative adjuvant radiotherapy and chemotherapy,concurrent radiotherapy and chemotherapy.Due to the specificity of esophageal anatomy and esophageal cancer biological characteristics,advanced esophageal cancer prone to regional lymph node metastasis,vascular infiltration,surgical treatment does not ensure clean lymph nodes,postoperative adjuvant treatment of the need to highlight,can be visible to the naked eye of subclinical lesions to kill.Traditional chemotherapy drugs on the treatment of esophageal cancer is not ideal,mainly based on DDP and5-FU or its derivatives program.Targeted drug such as gefitinib,erlotinib and more for the treatment of esophageal cancer are on the clinical study,lacking of evidence-based medical evidence.Immunotherapy is also approved for clinical use,because of its high price and inaccurate effect m many people discouraged.But whether it is traditional chemotherapy drugs or targeted drugs still can not avoid liver and kidney damage and severe bone marrow suppression,which greatly limits the clinical application of chemotherapy drugs,because the patient can not tolerate and also can not achieve the desired results.The development of new drugs for the treatment of esophageal cancer were in full swing,the researchers actively from the traditional Chinese medicine,the use of traditional Chinese medicine based on the motherland,looking for other Chinese herbal medicine which can be similar to paclitaxel that has a strong ability anti-cancer.Traditional Chinese medicine ginseng has a high health and medicinal value,with anti-tumor,anti-aging,anti-fatigue and many other effects.In recent years,ginseng ingredients have been isolated in a variety of ways,including ginsenosides,polysaccharides and peptides and so on.Ginsenoside(Ginsenoside,GS)is the main active ingredient of ginseng,there are more than 60 kinds of ginsenosides are isolated,the content take up about 4%.Ginsenoside Rg3 and protopanaxadiol Rh2 are most commonly used in tumor cell apoptosis studies,and various in vitro experiments have shown that they have high antitumor activity.Ginsenoside Rg5 is a newly isolated saponin monomer in recent years with a molecular weight of 767.01.At present,in vitro cell experiments have shown that it has inhibitory effect on growth and apoptosis of breast cancer and cervical cancer cells [3,4],but Rg5 has no report on the role of human esophageal cancer cells.Objective to explore the effect of rare ginsenoside Rg5 on human esophageal cancer Eca-109 cells,to observe its effect on the growth status of esophageal cancer cells and to detect its effect on cell cycle and apoptosis.Purpose:Method:This research through the cultivation of human esophageal cancer cell lines Eca109,using different concentrations of ginseng saponin Rg5 action at the cell,through the CCK 8 in enzyme standard instrument detection at the same time under different concentrations and the role of the same concentration of different time human esophageal cancer cells absorbance,through calculating its survival or inhibition rate,in order to judge the esophageal cancer cell growth.At the same time,the cell experiment was carried out based on the results of the above results,and the effect of different concentrations of Rg5 on the cycle of esophageal cancer cells and the apoptosis of the cell was investigated.Result:1.In a certain concentration range,Rg5 inhibited the proliferation of esophageal cancer Eca109 cells,and showed concentration and time dependence;2.The effect of Eca109 on esophageal cancer cell line 24 h after different concentration of Rg5 was different,and with the increase of concentration,the change of cell growth state was larger;3.The effect of different concentration of Rg5 on esophageal cancer Eca109 cells was significantly higher than that of control group(P <0.05),and the effect of Rg5 with50、100umol/L on esophageal cancer Eca109 cells was significantly higher than that of control group(P <0.05)There was no significant difference between the two groups(P> 0.05);4.The apoptosis rate of esophageal cancer Eca109 cells increased with the increase of Rg5 concentration at different concentrations of Rg5,There was a significant difference between the experimental group and the control group(P< 0.05)in the experimental group and the control group.Conclusion:In a certain concentration range,ginsenoside Rg5 on human esophageal cancer cells inhibit proliferation,can cause cell cycle retention,and can induce apoptosis,and showing a dose-dependent.This is a clinical basis for the clinical development of new drugs for the treatment of esophageal cancer,with potential clinical application value.
