Study On Therapeutic Effects And Treatment Mechanism Of Chengshijuanbitang On Adjuvant Arthritisrats

Aim:Investigate the therapeutic effects and treatment characteristics of Chengshijuanbitang(CSJBT)on adjuvant arthritis(AA)rats,to clarify the mechanism for the treatment of rheumatoid arthritis(RA)based on the humoral immunity response and cell immunity response indexes.Methods:Rats were randomly divided into six groups: control group,AA model group,GTW group(0.01 g/kg),CSJBT-L group(2.55 g/kg),CSJBT-M group(5.10 g/kg)and CSJBT-H group(10.20 g/kg).Except the normal group,rats were injected FCA into the right etapedes to establish the AA model.After treated with CSJBT by intragastric administration for 14 days,general index(arthritis score,hindpaw swelling,weight and organ indexes),hemodynamic indexes(whole blood viscosity,plasma viscosity and erythrocyte deposited),serum biochemical indices(immunoglobulin-G and immunoglobulin-M)and morphologic changes(liver,spleen,thymus and ankle)were measured to evaluate the effects of CSJBT on AA rats.PFC test was used to evaluate the change of humoral immunity in AA rats treated with CSJBT.MTT method was adopted to detect the changes in the splenic T and B-lymphocyte proliferation and the activity of NK cells.Effect of CSJBT to T-lymphocyte subset(CD3+CD4+?CD3+CD8+),NK(CD3–CD161+)and NKT(CD3+CD161+)cell in spleen of AA rat were detected by flow cytometer.Results:CSJBT promoted the weight recovery,relieved the paw swelling,reduced the arthritis score and Ig G level,and increased the liver index(CSJBT-M,P<0.05)of the AA rats significantly.Additionally,CSJBT decreased the whole blood viscosity and erythrocyte deposited,improved the pathological changes of liver,spleen and thymus,and inhibited the synovial hyperplasia and inflammatory cell invasion.Therefore,CSJBT has good therapeutic effects on AA rats.CSJBT reduced the number of plaque in PFC test,inhibited splenic T and B lymphocyte proliferation of AA rats and decreased the content of CD3+CD4+ T-lymphocytes and NK cells.MTT test showed that CSJBT inhibited the NK cell activity in spleen of AA rats.Conclusion:CSJBT has good therapeutic effects on AA rats,and can maintain the balance of CD4+/CD8+ and inhibit expression of NK cells which suggests the treatment mechanisms are related to regulating the immune function.
