Paeonol is an important active ingredient of traditional Chinese medicine,exists in Moutan Cortex and other Chinese herbal medicines,with a variety of pharmacological effects.Determination of the content of paeonol in Chinese herbal medicinal materials or Chinese patent medicine is of great significance to the quality evaluation of traditional Chinese medicine.In this thesis,the fluorescence enhancement reaction of paeonol with aluminum ions is studied,and the best experimental conditions are confirmed.On this basis,fluorometric method for the determination of paeonol in Moutan Cortex and several kinds of Chinese patent medicine is studied.And the reliability of the new method is verified by high performance liquid chromatography(HPLC).Three parts are included in the thesis:1.The fluorescence property and optimum experimental conditions of paeonol-Al(?)complex are studied.Paeonol itself has very weak fluorescence.In acidic conditions,adding aluminum ions will make its fluorescence significantly enhanced.The excitation wavelengths of the fluorescent complex are 350,290 and 235 nm,and the emission wavelength is 451 nm.Based on the fluorescence of paeonol-Al(?)complex,a fluorospectrophotometric method and a three-dimensional fluorescence coupled with second-order correction(3DF-2OC)method are employed respectively for the determination of paeonol in Moutan Cortex control drug and in commercial drug,the results of the two methods are basically the same.The contents of paeonol in the control drug and in the commercial drug are 1.26% and 2.05%,respectively.The results were verified by HPLC,which indicated that the fluorometric method is accurate and reliable.2.Based on the fluorescence property of paeonol-Al(?)complex,three kinds of Chinese patent medicine containing medicinal material Moutan Cortex are qualitatively and quantitatively analyzed.The composition of Chinese patent medicine is complex,the fluorescence peak of the coexisting component partially overlaps with the fluorescence peak of the paeonol-Al(?)complex,so,the content of paeonol can not be accurately determined by fluorospectrophotometric method,but the 3DF-2OC method can distinguish paeonol from the interference component according to the difference of fluorescence peak,and thus can get more accurate results.The method is used to determine the content of paeonol in Guizhi Fuling Capsule,Guizhi Fuling Pills and Liuweidihuang Capsule.The results obtained were 0.862%,0.225% and 1.48%,respectively.The results are verified by HPLC,and it is proved that the 3DF-2OC method is accurate.3.The contents of paeonol in three kinds of Chinese patent medicine containing medicinal material Xu Changqing are determined.These Chinese patent medicines contain other fluorescent components,and their fluorescence peaks overlap with the fluorescence peaks of the paeonol-Al(?)complex,so the contents of paeonol in these medicines can not be accurately determined by fluorospectrophotometric method.The 3DF-2OC method is used to determine the contents of paeonol in Rheumatism Capsule,Guci Pills and Bonesetting Liquid.The results are 0.463%,0.262% and 0.0245%,respectively.The results are validated by HPLC and proved to be accurate.
