Objective Research have shown that adipose-derived stem cells(ADSCs)have similar biological characteristics to bone marrow mesenchymal stem cells(BMSCs)in vitro,that is,they have a strong autologous Replication,cross-germination or cross-line differentiation ability,in the long-term in vitro culture process still maintain the ability of multi-directional differentiation,while the genetic background is stable,less rejection after transplantation and other advantages,and fat stem cells with convenient,self-To meet the needs and other advantages.It has become a hot research topic in recent years.At present,the method of obtaining human adipose-derived stem cells in clinical application is mainly to separate and culture the fat mixture obtained after liposuction by type ? collagenase digestion method.However,during the use of liposuction swelling anesthesia,And drug concentration on adipocytes and isolated culture of adipose derived stem cells have a negative impact on the biological characteristics,little known,rare at home and abroad related research reports.The adipose tissue of rats is relatively abundant,and the ethical problems and test items are limited,and it is a good test material.In this study,adipose tissue was extracted from adipose tissue and cultured adipose tissue,and the adipose tissue was extracted from rats.The adipose tissue was cultured and the effects of different concentrations of ropivacaine on rat fat Stem cell proliferation and adipogenic effects,to provide the basis for the best way to improve the anesthesia program in the process of adipose-derived stem cells,to obtain high-performance adipose-derived stem cells,reduce cell depletion,increase the utilization of adipose-derived stem cells,and use Its transplantation to repair skin and soft tissue defects and other aspects of orthopedic surgery to provide a theoretical basis.Materials and Methods 1)Design:Cytological in vitro observation.2)Material:From 4 weeks old rats with bilateral inguinal adipose tissue.3)Methods:Isolation and culture of adipose derived stem cells?Intervention of tumescent fluid?Detection of proliferation ability of adipose derived stem cells by CCK-8 kit and Brdu incorporation ?Adipogenic ability test.4)Outcome measures:?Morphological characteristics of rat adipose derived stem cells.?Multiplication capacityof adipose derived stem cells.?The rate of adipogenic differentiation.Results Primary cultured cells were larger spherical mononuclear cells,digested and passaged,the cell growth rate was significantly faster than the original cells,2 to 3 days to cover more than 85%.CCK-8 found that 25?mol / L,50?mol / L,100?mol / L ropivacaine treatment of adipose-derived stem cells 24 h,48h,72 h after the absorbance decreased significantly,compared with the control group,the difference was statistically significant.The results showed that ropivacaine could significantly reduce the proliferation ability of adipose-derived stem cells.At 24 h,the higher the concentration,the stronger the ability of suppressing stem cell growth.At 48 h and 72 h,the concentration and the inhibitory effect were not significantly different(44.61 ± 4.43)% in the control group,(46.30 ± 3.25)% in the 25 ?mol / L ropivacaine treatment group,49.46 ± 2.34% in the 50 ?mol / L ropivacaine treatment group,(50.07 ± 5.29)% in the 100 ?mol / L ropivacaine treatment group.The results showed that low dose ropivacaine had no significant effect on adipogenic ability of adipose-derived stem cells.After treatment with high concentration of ropivacaine,adipose stem cells Of the adipogenic capacity of the impact.When the concentration rose to a certain value,the fattening rate of adipose-derived stem cells did not change with the increase of concentration,but still need further test confirmed.Conclusions Ropivacaine treatment can significantly reduce the proliferation of adipose-derived stem cells.At 24 h,the higher the concentration,the stronger the ability of suppressing stem cell growth.At 48 h and 72 h,the concentration and the inhibitory effect were not significantly different.Low concentration of ropivacaine had no significant effect on adipogenic ability of adipose-derived stem cells,and high-concentration ropivacaine had an effect on the adipogenic ability of adipose-derived stem cells.When the concentration rose to a certain value,the fattening rate of adipose-derived stem cells did not change with the increase of concentration,but still need further test confirmed.
