Background and PurposeBreast cancer is the most common tumor in women.In recent years,the incidence of breast cancer has been increasing year by year.With the development of molecular biology technology,the occurrence and metastasis of tumor is a complicated process with many factors.It has been found that the human epidermal growth factor receptor(HER-2)is one of the most important factors affecting the prognosis of breast cancer.The patients with HER-2 gene amplification had the high recurrence and metastasis,shorter survival time,and poor prognosis in the clinical.The relationship between HER-2 genes and other molecular biological characteristics is the hotspot at home and abroad,The correlation between HER-2 gene amplification or overexpression and other indicators of molecular biology and tumor histology,HER-2 genes and other biological indicators such as tumor size,histological grade,lymph node metastasis,the expression of ER are quite different,even opposite results.It is different from the detection method,the evaluation criteria and the set of standard.It is difficult to accurately reflect the characteristics of the expression of HER-2 gene in breast cancer,which is not conducive to guide the clinical treatment.The study combined immunohistochemistry and fluorescence in situ hybridization of HER-2 gene status,strictly following the standard interpretation guidelines,study the relationship between overexpression of HER-2 gene amplification and clinical characteristics of patients with breast cancer and other clinical pathological characteristics.It is significance for the further understanding of the treatment and clinical expression of HER-2 gene in breast cancer.MethodsThere were 363 breast cancer patients who were treated in the Second Affiliated hospital of Zheng Zhou University from December 2013 to March 2017 in this study.All specimens were invasive ductal carcinoma of the breast by pathology.According to the immunohistochemical expression of HER-2 results.The uncertain results were detected by FISH.According to immunohistochemistry and FISH were divided into HER-2 expression group and no expression group,expression group including over expression of HER-2(+ + +),the positive expression of FISH;no expression grou including HER-2(-,+),FISH negative results.The clinical data such as age,tumor size and lymph node metastasis and other clinical data were collected.The study analyzed the clinical characteristics of HER-2 gene expression in breast cancer patients and the relationship between the two groups.We used the software of SPSS 17.0 to carry on the statistic analysis.Chi square test was used to analyzed the data.Correlation analysis was carried out by Spearman correlation analysis.Set ?=0.05 as a statistical test.Results1.Among 363 cases breast cancer patients,the samples of HER-2 negative was 304 cases(83.7%),HER-2 positive group was 59 cases(16.3%).2.The single factor analyzed showed that there was significant difference between the expression of HER-2 group and no expression in lymph node metastasis group(59.3%vs38.5%),the positive expression of ER(74.6%vs87.2%)and histological grade(27.2%vs26.9%;64.4%vs63.2%;8.5%vs9.8%),Ki-67?14%(91.5%vs74.0%)(p < 0.05).2.The correlation between the HER-2 expression group and no expression group showed that the lymph node metastasis,histological grade and Ki67 were positively correlated with HER-2(r1=0.28,r2=0.45,r3=0.34,p< 0.05),the expression of ER was negatively correlated(r1=-0.38,p< 0.05).Conclusions1.The expression of HER-2 is correlated with the lymph node metastasis?ER?the histological grade and the Ki67.
