Aims:To investigate the function and clinical significance of high-mobility group box-1(HMGB1)and toll-like receptors 4(TLR4)in temporal lobe intractable epilepsy.Methods:Nissl Staining were used to observe the morphology of nerve cells.Immunohistochemistry and western-blot were used to detect the expression of HMGB1 and TLR4 in 79 epileptogenic tissue of temporal lobe intractable epilepsy and 35 contrast brain tissue in Department of Neurosurgery of the Fifth Affiliated Hospital of Zhengzhou University from 2013-7-1 to 2015-11-30.The experiment date were analyzed statistically.Results:1.To observe the morphology of nerve cells by Nissl Staining,the number of nerve cells in epileptogenic tissue(197.14±33.55)was less than that in contrast brain tissue(249.16±52.26)and the difference was statistically significant(t=6.36,P <0.001).2.Immunohistochemistry showed the positive rates of HMGB1 protein in contrast brain tissue and epileptogenic tissue were 40%(14/35)and 86.08%(68/79);The expression of HMGB1 protein in epileptogenic tissue was higher than that in contrast brain tissue(?2=25.503,P<0.001);Western-blot showed the expression of HMGB1 protein in epileptogenic tissue was higher than that in contrast brain tissue(t=24.711,P<0.001)by comparing the optical density.The expression of HMGB1 protein in epileptogenic tissue had negative relation with gender(Z=-0.777,P>0.05)and age(Z=-0.777,P>0.05),but had positive relation with the medical history of epilepsy(Z=-2.758,P < 0.05),duration of epilepsy(Z=-2.431,P < 0.05),seizure frequency(Z=-3.294,P<0.05)and seizure type(Z=-3.532,P<0.001).3.Immunohistochemistry showed the positive rates of TLR4 protein in contrast brain tissue and epileptogenic tissue were 28.57%(10/35)and 71.43%(66/79);The expression of TLR4 protein in epileptogenic tissue was higher than that in contrast brain tissue(?2=32.984,P<0.001);Western-blot showed the expression of TLR4 protein in epileptogenic tissue was higher than that in contrast brain tissue(t=12.366,P<0.001)by comparing the optical density.The expression of HMGB1 protein in epileptogenic tissue had negative relation with gender(Z=-0.176,P>0.05)and age(Z=-0.259,P>0.05),but had positive relation with the medical history of epilepsy(Z=-2.103,P < 0.05),duration of epilepsy(Z=-2.103,P < 0.05),seizure frequency(Z=-2.779,P<0.05)and seizure type(Z=-2.151,P<0.001).4.The rank correlation analysis was taken between the expression of HMGB1 and TLR4 in epileptogenic tissue.HMGB1 and TLR4 were positively correlated in epileptogenic tissue(r=0.482,P<0.001).Conclusion:1.Inflammation caused by the long-term recurrent epileptogenesis can lead to neuronal damage.2.The expression of HMGB1 and TRL4 in epileptogenic tissue were higher than that in contrast brain tissue,and they were positively correlated in epileptogenic tissue.3.The activation of HMGB1/TLR4 signal pathway is significantly associated with temporal lobe intractable epilepsy and may provide a new target for treating temporal lobe intractable epilepsy.
